1 alpha,25-Dihydroxyvitamin D3 and 1 alpha-hydroxyvitamin D3 prolong survival time of mice inoculated with myeloid leukemia cells.

Abstract: Syngeneic SL mice inoculated with murine myeloid leukemia cells (Ml) all died ofleukemia within 30 days. Treatment three times a week with 12.5-50 pmol per mouse of either la,25-dihydroxyvitamin D3 [la,25(OH) It is of great interest that some tumor cell lines also possess similar cytosol receptors to which la,25(OH)2D3 binds specifically (4-6). However, the biological function of la,25(OH)2D3 in tumor cells was not known until quite recently. In 1981, Colston et aL (7) (17,18), the most potent known stimulator… Show more

“…To examine the effects of hypergravity on vitamin D-regulated gene expression, we treated mice with 1␣(OH)D 3 via intraperitoneal injection. 1␣(OH)D 3 is rapidly converted to 1,25(OH) 2 D 3 after injection and is more effective than 1,25(OH) 2 D 3 in prolonging survival time in a mouse model of leukemia (11). As reported previously (13,27), 1␣(OH)D 3 induced mRNA expression of VDR target genes, Cyp24a1, calbindin D9k, Trpv5, and Trpv6, in the kidney of ICR mice (Fig.…”

“…1,25-Dihydroxyvitamin D3 (calcitriol, 1,25D3) is well known to induce differentiation of leukaemic cells (Abe et al, 1981;Honma et al, 1983). This differentiation is linked to exit from the cell cycle at the restriction (R) point in late GI stage, resulting in growth inhi-bition, which suggests a new strategy for cancer therapy (Pardee, 1974).…”

“…1,25-dihydroxyvitamin D 3 , calcitriol, an active form of vitamin D 3 , has been reported to induce differentiation and inhibit the proliferation of various types of cancer cells in addition to its well-known biological activities in enhancing intestinal calcium transport and bone and mineral mobilization [2][3][4][5]. However, clinical application of this compound is limited because of its hypercalcemia-inducing activity.…”
