1,8-Cineole potentiates IRF3-mediated antiviral response in human stem cells and in an ex vivo model of rhinosinusitis

Abstract: The common cold is one of the most frequent human inflammatory diseases caused by viruses and can facilitate bacterial superinfections, resulting in sinusitis or pneumonia. The active ingredient of the drug Soledum, 1,8-cineole, is commonly applied for treating inflammatory diseases of the respiratory tract. However, the potential for 1,8-cineole to treat primary viral infections of the respiratory tract remains unclear. In the present study, we demonstrate for the first time that 1,8-cineole potentiates poly(… Show more

“…In this regard, most endogenous ligands studied until now seem to activate NF- κ B signalling via TLR4 [ 41 ], particularly fibrinogen [ 42 ] and HMGB1 [ 43 , 44 ]. U251 cells endogenously express proteins involved in the TLR4 signalling pathway and do not need to be transfected [ 25 , 26 ]. In U251-NF- κ B-GFP-Luc cells, fibrinogen had no effect on NF- κ B-activation whereas HMGB1 seemed to decrease the activity of NF- κ B.…”

“…Controls were treated within medium described above without additional stimuli. For gene expression analysis, treatments were performed for 4 h, while a 2 h treatment was done for immunocytochemistry in accordance to our previous studies [22], and a 5 h treatment for western blot.…”

“…In response to physical or physiological stress, injury, or infection, inflammation takes place as a key defense process of innate immunity aiming to restore the physiological situation. NF-κB is broadly described to be one of the key transcription factors regarding pro-inflammatory signaling, particularly activated by the presence of pro-inflammatory cytokines (like TNFα or IL-1), lipopolysaccharides (LPS) of the bacterial cell wall [ 17 ], or viral and bacterial nucleic acids [ 18 ]. Recognition of cytokines or LPS species is mediated by the respective receptors, such as TNF receptors or Toll-like microbial pattern recognition receptors (TLRs).…”
