1, 4-Dihydropyridines: A Class of Pharmacologically Important Molecules

Abstract: The 1, 4-dihydropyridines (DHPs), a class of drugs possess a wide variety of biological and pharmacological actions. It represents one of the most important groups of calcium-channel modulating agents and has experienced widespread use in the treatment of cardiovascular disease which includes antihypertensive, antianginal, vasodilator and cardiac depressants activities. It also shows antibacterial, anticancer, antileishmanial, anticoagulant, anticonvulsant, antitubercular, antioxidant, antiulcer, CFTR, antimal… Show more

“…Various modulators can potentiate or block calcium entry through L-type calcium channels. [4][5][6] Calcium antagonists have a versatile pharmacological activity such as antihypertensive and antianginal, [7][8][9] antitumor, 10,11) anti-inflammatory, 12,13) antitubercular, 14,15) anticonvulsant and antithrombotic. 16,17) 1,4-DHPs bind selectively to L-type calcium channel protein, precisely at the transmembrane domain IIIS6 and IVS6 regions of the α1 subunit.…”

“…1,4-DHPs possess several important pharmacological properties including calcium-channel-blocking, bronchodilator, anti-ischemic and antitumor as well as MDR reversal effects [10][11][12][13][14][15]. Several agents structurally related to 1,4-DHP derivatives have been reported as P-gp inhibitors [16][17][18][19][20][21].…”

“…The effects are expressed on smooth and cardiac muscle and even beyond the blood-brain barrier. In fact, some DHPs work better on certain types of vessels, for example on the coronary arteries or the cerebral vessels [5]. The striated muscles are free from these effects because the mechanism is a function of the calcium already present within the cells.…”

“…Currently, calcium antagonists used as antihypertensive agents are almost exclusively DHP compounds, while non-DHP drugs are mainly used as antiarrhythmics [5][6][7].…”
