1,3-Diaryltriazenes: a new class of anorectic agents

Abstract: A series of substituted 1,3-diaryltriazenes has been synthesized and tested for anorectic activity. Several members of the series were effective; one compound, 1,3-bis[2-cyano-5-(trifluoromethyl)phenyl]triazene, was particularly active, causing weight loss in rats, dogs, and squirrel monkeys. It was devoid of overt central nervous system activity, and compared to previously reported biologically active triazenes, it was relatively nontoxic up to 30 days of drug administration.

“…Several examples are provided in Table . The procedure enables the very convenient preparation of various 1,3-diaryltriazenes, a class of compounds from which representatives are known to possess anorectic activity . Some of them were recently used as starting materials for the preparation of 1,3-diaza-2-azoniaallene salts, novel N 3 -building blocks .…”

“…In industry, they can be used as chemical and biological reagents 13,14 and in organic synthesis [15][16][17] . The pharmaceutical industry has invested in studies on action against microorganisms 18,19 , functioning as appetite reducers 20,21 , anticancer agents [22][23][24][25][26] such as melanoma-specific 25 and primary brain tumors 28 therapeutics, fluorescence sensing agents 29 , mutagenic factors 12 , and having diabetic, anti-inflammatory, and antioxidant 27 activities; triazenes also exhibit biological and environmental activities 6,[30][31][32][33] .…”

“…Furthermore, this structure is capable of having a specific medicinal or antibacterial properties due to having two conformations of E and Z, which is can be investigated by other researchers in future. This structure in synthesized by a new method is investigated and characterized by FT-IR, Absorbance electron spectrum, 1 H-NMR, 13 C-NMR, and X-ray single crystal diffraction. The theoretical calculations for this structure are discussed in the following using DFT method in three levels of B3LYP, B3PW91, and PBEPBE.…”

“…In this study, it is tried to show a different and modern method for synthesizing these compounds.Triazene compounds have been used for the preparation of several sensors for determination of heavy metals, [9,10] and also used in solid-phase extraction. [11,12] They have been studied for their anorectic activity and potency against specific tumor cell lines, [13][14][15][16][17] as well as they have been applied as protecting groups in natural product synthesis [18,19] or used to form heterocycles. [20,21] Various compounds have been prepared from these compounds, because of linkage ability of them to carbonic nano tubes that have important abilities especially in man's body metabolism.…”
