1,3,5,7-Tetramethyl-8-(N-hydroxysuccinimidyl butyric ester)difluoroboradiaza-s-indacene as a new fluorescent labeling reagent for HPLC determination of amino acid neurotransmitters in the cerebral cortex of mice

Abstract: 1,3,5,7-Tetramethyl-8-(N-hydroxysuccinimidyl butyric ester)difluoroboradiaza-s-indacene (TMBB-Su), a new BODIPY-based fluorescent probe, was designed and synthesized for the labeling of amino compounds. It was used as a pre-column derivatizing reagent for determination of amino acid neurotransmitters by high-performance liquid chromatography (HPLC). The fluorescence quantum yield in acetonitrile increased from 0.84 to 0.95 when it reacted with amino acid neurotransmitters. Derivatization of TMBB-Su with seven … Show more

“…Numerous studies have demonstrated that the synthesis of proteins was inhibited, and the decomposition process was activated, which lead to increased amino acid levels in the blood [32,33,34,35]. Serine and alanine serve as fuel substrates in energy metabolism for brain cells to meet their high energy demands.…”

“…25 The use of N-acetyl aspartic acid (Naa) as an indicator of neuronal death originates from the early observation that its level is markedly lower in the cerebral infarction area in stroke. 26 Other AAs, such as alanine (Ala), 27 have been reported to be present at varying levels before and after cerebral ischemia, but their relationships with stroke and the mechanisms involved are not clear.…”
