1,25-Dihydroxyvitamin D3 and cisplatin synergistically induce apoptosis and cell cycle arrest in gastric cancer cells

Abstract: 1,25-Dihydroxyvitamin D3 [1,25(OH)2D3] plays an anticancer role in multiple types of cancer and potentiates the cytotoxic effects of several common chemotherapeutic agents. The hypercalcemia caused by 1,25(OH)2D3 alone or resistance to cisplatin weaken the anticancer effects of vitamin D. Thus, in this study, we aimed to investigate the synergistic effects of 1,25(OH)2D3 and cisplatin on the apoptosis and cell cycle progression of gastric cancer cells. BGC-823 human gastric cancer cells were treated with 1,25(… Show more

“…The ability of 1,25(OH) 2 D 3 to induce growth arrest has been widely studied in cancer cells, such as ovarian, prostate, breast, and gastric cancer cells [36][37][38][39], but this is the first report of this effect in human preadipocytes.…”

“…effects in inhibition of inflammatory markers' gene expression (17,29). Another proposed mechanism involves the roles of these vitamins in decreasing systemic inflammation, and subsequently the GC incidence, by reducing insulin resistance (40)(41)(42)(43). In case of Vitamin B6, as this vitamin has a crucial role in amino acid and amines metabolism, it is logical to assume an essential part for this vitamin in reducing the chronic inflammation.…”

“…Vitamin D 3 may act as an antagonist of hedgehog signaling to suppress viability of gastric cancer cells, and it also has a synergistic effect with other anticancer drugs by reducing mRNA expression of the target genes of hedgehog signaling ( Ptch1, Gli1, cyclin D1 and bcl2) [95]. Co-treatment with cisplatin and 1α,25(OH) 2 D 3 enhanced cisplatin-mediated cell growth inhibition and cell apoptosis of human gastric cancer cells with an upregulation of Bax, a decrease in ERK and AKT phosphorylation levels, and an increase in p21 and p27 levels [96]. …”
