The genetic determination of eggshell coloration has not been determined in birds. Here we report that the blue eggshell is caused by an EAV-HP insertion that promotes the expression of SLCO1B3 gene in the uterus (shell gland) of the oviduct in chicken. In this study, the genetic map location of the blue eggshell gene was refined by linkage analysis in an F2 chicken population, and four candidate genes within the refined interval were subsequently tested for their expression levels in the shell gland of the uterus from blue-shelled and non-blue-shelled hens. SLCO1B3 gene was found to be the only one expressed in the uterus of blue-shelled hens but not in that of non-blue-shelled hens. Results from a pyrosequencing analysis showed that only the allele of SLCO1B3 from blue-shelled chickens was expressed in the uterus of heterozygous hens (O*LC/O*N). SLCO1B3 gene belongs to the organic anion transporting polypeptide (OATP) family; and the OATPs, functioning as membrane transporters, have been reported for the transportation of amphipathic organic compounds, including bile salt in mammals. We subsequently resequenced the whole genomic region of SLCO1B3 and discovered an EAV-HP insertion in the 5′ flanking region of SLCO1B3. The EAV-HP insertion was found closely associated with blue eggshell phenotype following complete Mendelian segregation. In situ hybridization also demonstrated that the blue eggshell is associated with ectopic expression of SLCO1B3 in shell glands of uterus. Our finding strongly suggests that the EAV-HP insertion is the causative mutation for the blue eggshell phenotype. The insertion was also found in another Chinese blue-shelled breed and an American blue-shelled breed. In addition, we found that the insertion site in the blue-shelled chickens from Araucana is different from that in Chinese breeds, which implied independent integration events in the blue-shelled chickens from the two continents, providing a parallel evolutionary example at the molecular level.
Chicken prolactin (PRL) is a physiological candidate gene for egg production. The objective of the current research was to investigate the association of polymorphisms in the chicken PRL promoter region with egg production. Genotyping of 177 individuals from White Leghorn, Yangshan, Taihe Silkies, White Rock, and Nongdahe breeds for 6 single nucleotide polymorphisms (C-2402T, C-2161G, T-2101G, C-2062G, T-2054A, and G-2040A) and 1 24-bp indel (insertion-deletion) at the site of -358 of the chicken PRL gene revealed large breed differences in allelic frequencies for all but the T-2101G and T-2054A polymorphisms. An F2 population produced from Nongdahe x Taihe Silkies chickens consisted of 374 hens, which were recorded for egg production traits and genotyped for the above 7 polymorphisms. Marker-trait association analysis indicated that the 24-bp indel was associated with egg production (P < 0.01) and that H3 (C C T C T G) was the most advantageous haplotype for egg production.
Domestic sheep and their wild relatives harbor substantial genetic variants that can form the backbone of molecular breeding, but their genome landscapes remain understudied. Here, we present a comprehensive genome resource for wild ovine species, landraces and improved breeds of domestic sheep, comprising high-coverage (∼16.10×) whole-genomes of 810 samples from 7 wild species and 158 diverse domestic populations. We detected, in total, ∼121.2 million single nucleotide polymorphisms (SNPs), ∼61 million of which are novel. Some display significant (P < 0.001) differences in frequency between wild and domestic species, or are private to continent-wide or individual sheep populations. Retained or introgressed wild gene variants in domestic populations have contributed to local adaptation, such as the variation in the HBB associated with plateau adaptation. We identified novel and previously reported targets of selection on morphological and agronomic traits such as stature, horn, tail configuration and wool fineness. We explored the genetic basis of wool fineness and unveiled a novel mutation (chr25: T7068586C) in the 3’-UTR of IRF2BP2 as plausible causal variant for fleece fiber diameter. We reconstructed pre-historical migrations from the Near Eastern domestication center to South-and-Southeast Asia, and found two main waves of migrations across the Eurasian Steppe and the Iranian Plateau in the Early and Late Bronze Ages. Our findings refine our understanding of genome variation as shaped by continental migrations, introgression, adaptation and selection of sheep.
Based on the knowledge of the heme bio-synthetic and metabolic pathway and the structures of biliverdin and protoporphyrin, experiments were carried out to compare the difference between the total quality of eggshell pigments in blue-shelled eggs and brown-shelled eggs from the same population (Dongxiang, China) and to analyze the correlation between the quantity of protoporphyrin and biliverdin in the 2 kinds of eggshells. It was found that there was no significant difference between the total quantity of eggshell pigments in Dongxiang blue-shelled eggs and Dongxiang brown-shelled eggs (P = 0.9006), and a highly significant positive correlation between the quantity of protoporphyrin and biliverdin in blue eggshells (P < 0.01) and a significant positive correlation between the quantity of protoporphyrin and biliverdin in brown eggshells (P < 0.05). These results suggested that eggshell protoporphyrin and eggshell biliverdin probably derived from common precursor material.
To investigate the value of the combined detection of the neutrophil-to-lymphocyte ratio (NLR) and Creactive protein level (CRP) in the diagnosis of COVID-19. A total of 191 patients with COVID-19 were recruited at the Third Hospital of Wuhan from 21 January 2020 to 20 February 2020. Fifty healthy volunteers were randomly selected as the control group. Age, gender, white blood cell count (WBC), CRP, lymphocyte percentage, and NLR were extracted. Quantitative clinical characteristics and laboratory values were compared between groups. Risk factors and receiver operating characteristic (ROC) curves for COVID-19 were analyzed. We found that the NLR and CRP were higher, while the lymphocyte percentage was lower in patients with COVID-19 than in healthy controls. Among patients confirmed to have COVID-19, the NLR and CRP of the moderate group were lower than those of severely ill patients (severe, critical and death groups), and the lymphocyte percentage of the moderate group was higher than that of the critical and death group. There were no significant differences in WBC among all groups. Logistic regression analysis showed that the NLR, CRP, and lymphocyte percentage were independent risk factors for COVID-19. The AUC of the combined determination of NLR and CRP was 0.863, which was higher than that of NLR, CRP, WBC, and lymphocyte percentage (AUC: 0.835, 0.775, 0.416, and 0.749, respectively). Our results showed that the NLR and CRP were independent risk factors for COVID-19, and the combined detection of the NLR and CRP showed improved diagnostic performance for COVID-19.
