Cationic niosomes have become important non-viral vehicles for transporting a good number of small drug molecules and macromolecules. Growing interest shown by these colloidal nanoparticles in therapy is determined by their structural similarities to liposomes. Cationic niosomes are usually obtained from the self-assembly of non-ionic surfactant molecules. This process can be governed not only by the nature of such surfactants but also by others factors like the presence of additives, formulation preparation and properties of the encapsulated hydrophobic or hydrophilic molecules. This review is aimed at providing recent information for using cationic niosomes for gene delivery purposes with particular emphasis on improving the transportation of antisense oligonucleotides (ASOs), small interference RNAs (siRNAs), aptamers and plasmids (pDNA).

Stroke represents an attractive target for stem cell therapy. Although different types of cells have been employed in animal models, a direct comparison between cell sources has not been performed. The aim of our study was to assess the effect of human multipotent adult progenitor cells (hMAPCs) and human mesenchymal stem cells (hMSCs) on endogenous neurogenesis, angiogenesis and inflammation following stroke. BALB/Ca-RAG 2−/− γC−/− mice subjected to FeCl3 thrombosis mediated stroke were intracranially injected with 2×105 hMAPCs or hMSCs 2 days after stroke and followed for up to 28 days. We could not detect long-term engraftment of either cell population. However, in comparison with PBS-treated animals, hMSC and hMAPC grafted animals demonstrated significantly decreased loss of brain tissue. This was associated with increased angiogenesis, diminished inflammation and a glial-scar inhibitory effect. Moreover, enhanced proliferation of cells in the subventricular zone (SVZ) and survival of newly generated neuroblasts was observed. Interestingly, these neuroprotective effects were more pronounced in the group of animals treated with hMAPCs in comparison with hMSCs. Our results establish cell therapy with hMAPCs and hMSCs as a promising strategy for the treatment of stroke.

Objetivos. Describir los hallazgos de un año de la vigilancia epidemiológica de diabetes mellitus en hospitales piloto pertenecientes al sistema de vigilancia de diabetes (VD). Materiales y métodos. Estudio transversal que incluyó a los pacientes diabéticos notificados durante el 2012 en 18 hospitales piloto del Perú. Se revisó la base de datos de la VD, obteniéndose variables epidemiológicas y de laboratorio como edad, sexo, tipo de diabetes, complicaciones, comorbilidad, microalbuminuria, glucosa en ayunas y hemoglobina glucosilada (HbA1c) al momento de captación por la VD y en el último control. Resultados. Se encontraron 2959 casos. Al momento de captación por la VD, 91,2% contaba con glicemia en ayunas de los cuales, 65,4% presentaba valor ≥ 130 mg/dL. El 8,9% contaba con estudio de microalbuminuria, el 20,5% de ellos presentaba microalbuminuria y el 6,5% proteinuria. En total, 1025 pacientes tuvieron consulta de control; 93,1% contaba con glicemia en ayunas y 22,3% con HbA1c. El 63,5% tenía glicemia en ayunas ≥ 130 mg/dL y 73,4% HbA1c ≥ 7,0%. La complicación más frecuente fue la neuropatía (21,4%) y la comorbilidad más frecuente la hipertensión arterial (10,5%). Se observó casos de tuberculosis y cáncer, de los cánceres, el más frecuente fue el de mama, principalmente en mujeres posmenopáusicas. Conclusiones. La VD muestra que en los diabéticos de los hospitales piloto en los que se cuenta con resultados de laboratorio existe una alta frecuencia control glicémico inadecuado y pobre adherencia al tratamiento. La alta frecuencia de complicaciones encontrada evidencia la necesidad de fortalecer el diagnóstico temprano.

Objetivos. Determinar la prevalencia de hipertensión y diabetes en habitantes de los distritos de Lima metropolitana y El Callao en Perú. Materiales y métodos. Estudio transversal analítico, realizado durante el mes de septiembre del 2006 en habitantes de 15 años de edad a más, residentes en Lima metropolitana y El Callao. Los participantes fueron seleccionados mediante un muestreo por conglomerados en tres etapas. Se utilizaron procedimientos estandarizados para medir peso, talla, perímetro de cintura, presión arterial y niveles de glucosa en sangre. Se realizó análisis univariados, bivariados y procedimientos de regresión logística para estimar los odds ratio (OR) y sus respectivos intervalos de confianza al 95%.

PURPOSE. The purpose of this study was to develop a culture method for expansion of corneal endothelial cells (CEC) based on the combined activation of PI3K/Akt and Smad2.METHODS. Morphology, proliferation, and migration of cultured rabbit and nonhuman primate CEC were examined in the presence of the PI3K/Akt activators IGF-1 and heregulin beta in combination with the Smad2 activator activin A. Phenotypic characterization of CEC was performed at the RNA and protein levels. Cell pump function and transepithelial electric resistance were used for in vitro functional assessment of CEC. Finally, ex vivo-expanded rabbit CEC were transplanted into a model of endothelial damage in rabbit corneas.RESULTS. Treatment of rabbit and nonhuman primate CEC in vitro with IGF-1, heregulin beta, and activin A induced an upregulation of PI3K/Akt and Smad2 signaling pathways and an increase in proliferation and migration of CEC expressing ZO-1, connexin-43, and Na þ /K þ -ATPase. Cell pump function evaluation revealed the complete functionality of cultured CEC. Injection of rabbit CEC successfully produced recovery of normal corneal thickness in a rabbit model of endothelial dysfunction.CONCLUSIONS. We demonstrated that the combined activation of PI3K/Akt and Smad2 results in in vitro expansion of phenotypic and functional CEC. Expanded cells were able to contribute to restoration of corneal endothelium in a rabbit model. These findings may represent a new therapeutic approach for treating corneal endothelial diseases.

Objetivo: Determinar la prevalencia del pie en riesgo de ulceración (PRU) según International Working Group on Diabetic Foot (IWGDF) expresada por la presencia de neuropatía diabética periférica (NPD) y/o enfermedad arterial periférica (EAP) y/o deformidad (D) y sus factores asociados en diabéticos tipo 2 en la atención primaria de la salud (APS) de Trujillo. Materiales y métodos: Se captó una muestra de 301 diabéticos tipo 2 de centros de salud, públicos y privados del distrito de Trujillo, pertenecientes a la APS; posteriormente se estratificó según el sistema IWGDF para pie en riesgo, se consideraron diagnósticos de NPD, EAP y D. Se realizó estadística descriptiva y analítica con las variables evaluadas por SPSS versión 22. Además, se solicitó una acreditación de un comité de ética e investigación. Resultados: Los diabéticos tipo 2 mostraron los siguientes diagnósticos: el 13,3 % presentaron NPD, EAP en 18,6 % y D en 64,1 %. Además, se encontró que el 86,7 % no presentan PRU, el 4 % se encuentra en un riesgo leve y el 9,3 % en moderado; con una prevalencia total de 13,3 %. Finalmente, se encontraron como factores de riesgo asociados a la regularidad de atención, hipertensión arterial, hipoglicemia, actividad física y calzado ortopédico. Conclusiones: Estudio epidemiológico único en la APS peruana que muestra una prevalencia de nivel moderado en PRU, pero menor respecto a sus similares latinoamericanos, siendo el componente deformidad la más frecuentes; y con factores asociados positivos y muy débiles cuando se analizó los pacientes con y sin riesgo. Moderate prevalence of foot ulceration risk according to the IWGDF guidelines in type 2 diabetic patients attending the primary health care ABSTRACT Objective: To determine the prevalence of foot ulceration risk (FUR) according to the International Working Group on the Diabetic Foot (IWGDF) guidelines, demonstrated by the presence of peripheral diabetic neuropathy (PDN) and/or peripheral arterial disease (PAD) and/or deformity (D), as well as its associated factors, in type 2 diabetic patients attending the primary health care (PHC) in Trujillo. Materials and methods: A sample of 301 type 2 diabetic patients was collected from public and private health centers of the district of Trujillo belonging to the PHC. Subsequently, said sample was stratified according to the IWGDF guidelines on foot at risk, considering diagnoses such as PDN, PAD and D. Descriptive and analytical statistics were performed with the variables evaluated by the IBM SPSS Statistics V22.0. In addition, accreditation was requested from an ethics and research committee. Results: Type 2 diabetic patients showed the following diagnoses: 13.3 % presented PDN, 18.6 % PAD and 64.1 % D.
