Although wheat and maize are the major economically important cereal crops and staple food sources in the world, their productivity is highly affected by excess salts in soil (salinity). Applications of multifarious halophilic plant growth-promoting rhizobacteria (PGPR) in saline soil protect the plants from osmotic damages and promote plant growth through the secretion of plant growth promoting (PGP) and osmolytes. In this study, Klebsiella variicola SURYA6—a PGPR—was evaluated for plant-growth-promotion and salinity amelioration in wheat and maize, and enrichment of soil nutrients. The results of the present study revealed that K. variicola SURYA6 grows luxuriously under high salinity stress conditions and produces copious amounts of three principal salinity ameliorating traits, such as 1 aminocyclopropane-1-carboxylate deaminase (ACCD), indole-3-acetic acid (IAA), exopolysaccharides (EPS), and osmolytes—such as proline, sugars, proteins, and amino acids. The isolate also exhibited sensitivity to a wide range of antibiotics, lack of hemolytic ability, and absence of catalase and oxidase activities confirming its nonpathogenic nature. Inoculation of wheat and maize seeds with this multifarious strain, improved the physicochemical properties of soil, improved seed germination by 33.9% and 36.0%, root length by 111.0%, 35.1%, shoot height by 64.8% and 78.9%, and chlorophyll content by 68.4% and 66.7% in wheat and maize seedlings, respectively, at 45 days after sowing (DAS) under salinity stress. The improvement in plant growth can be correlated with the secretion of PGP traits and improved, uptake of minerals such as nitrogen (N), phosphorus (P), sodium (Na), potassium (K), and magnesium (Mg). While amelioration of salinity can be the result of secretion of osmolytes and the change in pH from salinity to neutrality. This inoculation also significantly improved the soil nutrients under salinity stress conditions. Inoculation of K. variicola SURYA6, resulted in more improved growth and nutrients contents in plants and enriched soil nutrients under salinity stress as compared to normal (non-saline) conditions. Such multifarious strain can serve as a potent bio-inoculant for growth promotion of wheat and maize in saline soil. However, multi-year field trials under different agro-climatic conditions are required to confirm the bio-efficacy of K. variicola SURYA6.

This study aimed to isolate and identify moulds from rotten Citrus sinensis post-harvests and to investigate the activity of antagonist and biocontrol activity moulds that cause citrus fruit rotting. A total of 12 mould isolates were obtained. Following the pathogenicity test, two representative mould isolates were selected and identified based on the sequence analyses of internal transcribed spacer (ITS) regions of the rDNA. Methods used in this study include isolation of fungal postharvest diseases, pathogenicity assay, antagonism assay, growth curve analysis, in vitro biocontrol assay, and molecular phylogenetic analysis. Two isolates of fungal postharvest diseases were determined as the most destructive pathogens. The biocontrol assay showed that isolates of Y1 and Y10 were capable to reduce the growth of fungal isolates K6 and K9 and mitigate up to 100% of the damage of sweet citrus fruits after 7 days of incubation. The moulds were identified as K6 (Aspergillus flavus sensu lato) and K9 (Aspergillus niger sensu lato). Phylogenetic analysis showed that the Y10 yeast isolate was identified as Candida orthopsilosis, whereas the Y1 isolate had a close genetic relationship with Aureobasidium pullulans and possibly belongs to a new species. Further analysis is necessary to confirm this finding.

This study was focused on the effects of fermentation temperature and pH on the quality of Punica granatum juice probioticated with Lactobacillus species: Lactobacillus plantarum, Lactobacillus casei, Lactobacillus bulgaricus, and Lactobacillus salivarius. The whole fruit juice of P. granatum which is rich with phytonutrients appeared to be a good probiotic carrier. The probiotication was carried out for 24 hr at 30, 35, and 37°C and pH 2.5, 4.0, and 5.5 under microaerophilic conditions. The results found that P. granatum juice cultivated with L. casei had a better growth profile with a higher biomass density at 37°C around pH 3.5–4.0. Probiotication could maintain the scavenging activity of P. granatum juice cultivated with L. casei. The scavenging activity achieved up to 90% inhibition at the concentration of 5 mg/ml. The whole fruit‐squeezed P. granatum juice was suitable for the growth of Lactobacillus species even without supplementation during cultivation. Practical applications The findings of this study presented the potential of P. granatum juice (whole fruit) to be used as a good probiotic carrier, particularly for Lactobacillus species without supplementation. High nutritious P. granatum juice catered the need of probiotic bacteria during fermentation. Probiotication could maintain the antioxidant capacity of the juice in term of its radical scavenging activity. The antioxidant capacity was mainly attributed to the metabolites such as phenolic acids (romarinic acid and caftaric acid) and flavonoids (quercetin, quercetin 3‐glucoside, rutin and kaempferol rutinoside). With the optimized temperature (37°C) and pH (4.00), probiotic bacteria could growth well up to a cell viability of 2.46 × 1010 cfu/ml. This offers P. granatum to be developed into functional food to cater to the needs of the consumers who are lactose intolerant to dairy products.

This study discusses the isolation and identification of a new Streptomycetes highly active chitinase producer. Fifteen strains were isolated from Malaysian soil samples. The isolate WICC-A03 was found to be the most active chitinase producer. Its antifungal activity was evaluated against many phytopathogens. The identification of WICC-A03 using phenotypic and genotypic methods strongly indicated that strain WICC-A03 belonged to the genus Streptomyces and displayed similarity (91%) with Streptomyces glauciniger. Thus, it was given the suggested name S. glauciniger WICC-A03 with accession number: JX139754. WICC-A03 produces extracellular chitinase in a medium containing 1.5% colloidal chitin in submerged culture on 144 h. The produced enzyme was partially characterised and its molecular weight of 50 kDa was determined by using SDS-PAGE. This study indicates that WICC-A03 is a potential chitinase producer for biocontrol of plant pathogens. Further experiments are being carried out to optimise medium composition and cultivation conditions under lab and bioreactor scale.

The present study involves isolation of Streptomyces spp. from rhizosphere of Coscinium fenestratum Gaertn, an endangered medicinal plant from Western Ghats of Karnataka, India. Four potential isolates were identified by 16S rRNA sequencing as Streptomyces sp. RHPR3, Streptomyces puniceus RHPR9, Streptomyces sp. RHPR14 and Streptomyces mediolani RHPR25. An enrichment culture method was used for the isolation of Streptomyces spp. for biosurfactant activity. Among four potential Streptomyces spp., S. puniceus RHPR9 showed highest Emulsification index (EI) (78±0.2%) and Emulsification assay (EA) (223±0.2 EU mL-1). Thin layer chromatography, Fourier transform infrared spectroscopy (FTIR) and mass spectrometric analysis revealed that as glycolipid. Further confirmed by presence of fatty acids like hexanoic acid methyl ester, decanoic acid by Gas chromatography mass spectroscopy (GC-MS) analysis. S. puniceus RHPR9 showed a significant IAA production (41μg mL-1), solubilized P (749.1 μg mL-1), growth promotion of chilli (Capsicum annuum L.) was evaluated using paper towel method and greenhouse conditions. S. puniceus RHPR9 showed a significant increase in seed vigor index (2047) and increase in plant biomass (65%) when compared to uninoculated control. To our knowledge, this is the first report on epiphytic S. puniceus RHPR9 isolated from an endangered medicinal plant C. fenestratum Gaertn, for biosurfactant production and plant growth promotion activities.
