Uptake of nonesterified long-chain fatty acids (LCFAs) into many cell types and organs such as liver, heart, intestine, and skeletal muscle occurs primarily through a saturable, protein-mediated mechanism. Membrane proteins that increase the uptake of LCFAs, such as FAT/CD36 and fatty acid transport proteins, represent significant therapeutic targets for the treatment of metabolic disorders, including type 2 diabetes. However, currently available methods for the quantification of LCFA uptake neither allow for real-time measurements of uptake kinetics nor are ideally suited for the development of LCFA uptake inhibitors in high-throughput screens. To address both problems, we developed a LCFA uptake assay using a fluorescently labeled fatty acid and a nontoxic cell-impermeable quenching agent that allows fatty acid transport to be measured in real time using fluorescence plate readers or standard fluorescence microscopy. Using the components of a particularly well-studied plant virus, cowpea chlorotic mottle virus (CCMV), we demonstrate the synthesis of virus-like particles (VLPs) with one end of the packaged RNA extending out of the capsid and into the surrounding solution. This construct breaks the otherwise perfect symmetry of the capsid and provides a straightforward route for monofunctionalizing VLPs using the principles of DNA nanotechnology. It also allows physical manipulation of the packaged RNA, a previously inaccessible part of the viral architecture. Our synthesis does not involve covalent chemistry of any kind; rather, we trigger capsid assembly on a scaffold of viral RNA that is hybridized at one end to a complementary DNA strand. Interaction of CCMV capsid protein with this RNA-DNA template leads to selective packaging of the RNA portion into a well-formed capsid, but leaves the hybridized portion poking out of the capsid through a small hole. We show that the RNA-DNA protruding from the capsid is capable of binding DNA and DNA-functionalized colloidal particles. Additionally, we show that the RNA-DNA scaffold can be used to nucleate virus formation on a DNA-functionalized surface. We believe this self-assembly strategy can be adapted to viruses other than CCMV.

of DNA. Typically, acetylation of histone tails leads to gene activation associated with a loosely folded chromatin structure. Conversely, tri-methylation of histone tails and methylation of DNA lead to gene repression associated with a compacted structure of the chromatin. Despite decades-long efforts, the microscopic mechanism of epigenetic marker-mediated modulation of chromatin structure remains unknown. A possible explanation is the action of ''effector'' proteins that read epigenetic markers and alter DNA-DNA interactions depending on the message carried by the markers. However, none of the ''effector'' protein candidates characterized thus far can account for the observed chromatin compaction phenomena. Here, we combine molecular dynamics simulations with single-molecule FRET experiments to demonstrate modulation of DNA-DNA interactions by epigenetic markers that does not involving any of the ''effector'' proteins. In our MD simulations, we found that tri-methylations of histone tails and methylation of DNA cooperatively strengthen the histone-tail-mediated DNA-DNA association by increasing the number of lysine residues bridging the DNA molecules. Single-molecule FRET experiments confirmed the predictions of the molecular dynamics simulations. Our effector-free chromatin condensation mechanism suggests that epigenetic markers not only serve as information carriers but also can physically affect interactions between the DNA molecules.
