An unusual type I polyketide defined in a unique hexacyclic skeleton was determined as the structure of hexacyclinic acid (1). The structure was elucidated by X‐ray analysis in connection with chemical–spectroscopic methods, and the biosynthesis was explored through feeding experiments with 13C‐labeled precursors. The compound was isolated by application of the OSMAC approach to Streptomyces cellulosae S 1013.
The biosynthesis of the gabosines A−C (3−5) was studied by feeding cultures of Streptomyces cellulosae subsp. griseorubiginosus (strain S 1096) with 13C‐labeled precursors. Although the carba sugars are structurally related to shikimic acid, the biosynthetic origin was found to be different to the shikimate pathway. The results revealed that the gabosines are formed via a pentose phosphate pathway by cyclization of a heptulose phosphate intermediate. This intermediate arises from a triose phosphate by successive transfer of two C2 fragments by transketolases. This pathway is identical as that described for valienamine (7), the aminocarba sugar moiety of validamycin, and acarbose (2). The results from biosynthetic studies are discussed on the background of the variety of gabosines found in nature.
Wie aus früheren Untersuchungen bekannt, bildet Streptomyces cellulosae subsp. griseorubiginosus (Stamm S 1013) in Schüttelkulturen mit einem Haferflockenmedium lediglich die Carbazucker Gabosin D, 1, und E, 2. [1] Mit Hilfe des OSMAC-Ansatzes (OSMAC one strain/many compounds) [2] und des chemischen Screenings [3] sollte untersucht werden, ob sich bei diesem Stamm die Produktion neuer Metabolite induzieren lässt.Der OSMAC-Ansatz geht von der Beobachtung aus, dass einzelne Sekundärstoffproduzenten mehr Metabolite bilden können, als sich bei der Untersuchung von Standard-Rohextrakten erkennen lässt. [2] So ist es bei den für die Sekundärstoffbildung besonders begabten Actinomyceten und Pilzen leicht, auch bei gut untersuchten Stämmen neue Naturstoffe zu entdecken, deren Biosynthese letztlich durch eine Variation der Kultivierungsparameter induziert oder gefördert wird. Dabei gibt es einen flieûenden Übergang zwischen der Ausbeutesteigerung einzelner im Prinzip vorhandener und dem erstmaligen Auftreten vorher nicht nachweisbarer Metabolite. Wirkungsvoll variieren lassen sich beim OSMAC-Ansatz neben Nährmedium und Kultivierungsgefäû unter anderem Temperatur, Belüftung, pH-Wert und/oder Belichtung. Im Nährmedium spielen die Auswahl der C-und der N-Quelle sowie die Zugabe von anorganischen Salzen, Enzyminhibitoren oder Assemblierung von ApophyA65 mit 4 und Belichtung: Rekombinantes HaferApophyA65 [11] (Rohlysat aus Hansenula-polymorpha-Hefezellen; [11a] 4.5 nmol in 600 mL Phosphatpuffer, pH 7.6) wurde mit 4 (5 nmol in 1 mL DMSO) bei Raumtemperatur inkubiert. Die Apoprotein-Konzentration wurde separat vor der Inkubation durch Assemblierung eines Protein-Aliquots mit einem Überschuss an 2 bestimmt.Die Lösung des ApophyA65/4-Komplexes wurde mit dem Licht eines Diaprojektors bestrahlt (250 W; Abstand der Probe vom Projektor ca. 20 cm; die Anregungswellenlängen von l 614 AE 7 und 714 AE 7 nm für P r 3P fr bzw. P fr 3P r wurden mit Hilfe von Interferenzfiltern erzeugt). Für vollständige Umsätze (maximale Absorptionswerte) der jeweiligen Form waren Belichtungszeiten von ! 10 min erforderlich. Die Absorptionsspektren wurden mit einem Shimadzu-UV2102PC-Spektrometer aufgenommen.Verdrängungsexperiment mit ApophyA65/4-Komplex und Phycocyanobilin 2: Zu einer Probe (600 mL) der P r -ähnlichen Form des ApophyA65/4-Komplexes wurde eine Lösung von 2 (1.7 nmol in 1 mL DMSO) hinzugegeben. Die Mischung wurde für einige Minuten im Dunkeln stehen gelassen, bevor erst mit Licht der Wellenlänge 653 AE 7 nm (Interferenzfilter; P r 3P fr ) und darauf mit b 720 nm (Kantenfilter; P fr 3P r ) bestrahlt wurde. Vor der ersten und darauf nach jeder Belichtung wurden UV/Vis-Spektren aufgenommen.
