Biostimulants have been found effective in enhancing plant resistance toward stressful conditions. The aim of the present study was to evaluate the efficacy of selected biostimulants to overcome the negative effects of nutrient limitation on the growth performances and on the fruit quality of soilless cultivated strawberry plants. The condition of nutrient limitation was imposed by supplying the plants with only a single fertilization at transplantation and by excluding any further nutrient supply for the entire duration of the experiment (three months, from May to July). Strawberry plants were treated seven times during the period from preflowering up to berry maturation with different classes of biostimulants (humic acids, alfalfa hydrolysate, macroseaweed extract and microalga hydrolysate, amino acids alone or in combination with zinc, B-group vitamins, chitosan, and a commercial product containing silicon) at commercial dosages. The use of alfalfa hydrolysate, vitamins, chitosan, and silicon was able to promote biomass accumulation in roots (four to seven folds) and fruits (+20%) of treated plants, whereas the total leaf area increased by 15%–30%. Nutrient concentrations in leaves and roots showed variations for microelements (e.g., Fe, B, Zn, and Si) in response to biostimulant applications, whereas no significant differences were observed for macronutrient contents among treatments. Final berry yield was found around 20% higher in chitosan- and silicon-treated plants. Chitosan treatment significantly increased pulp firmness (by 20%), while a high nutritional value (e.g., phenolic compounds concentration) was observed in alfalfa- and seaweed-treated fruits (+18%–20% as compared to control). The overall outcomes of the present experiment show that selected biostimulants can be considered as a valid agronomic tool able to contrast the negative consequence of growing crops under insufficient nutritional conditions.
The tetrapyrrolic chlorophyll catabolites (or phyllobilins, PBs) were analyzed in yellow fall leaves of the grape Chardonnay, a common Vitis vinifera white wine cultivar. The major fractions in leaf extracts of V. vinifera, tentatively assigned to PBs, were isolated and their structures elucidated. The dominant fraction is a dioxobilin‐type non‐fluorescent Chl‐catabolite of a previously observed type. Two less polar fluorescent PBs were characterized as a novel dioxobilin‐type fluorescent Chl‐catabolite with a bicyclo‐1′,6′‐glycosyl architecture, and its new fluorescent formyloxobilin‐type analogue. The discovery of persistent hypermodified fluorescent PBs with the architecture of bicyclo‐[17.3.1]‐PBs (bcPBs), suggests the activity of an unknown enzyme that forges the 20‐membered macroring at the tetrapyrrolic core of a fluorescent PB. bcPBs may play specific physiological roles in grapevine plants and represent endogenous anti‐infective agents, as found similarly for other organic bicyclo‐[n.3.1]‐1′,6′‐glycosyl derivatives.
The experiment was conducted during two consecutive seasons (years 2016 and 2017) in an organic apple orchard of the cultivar Jonathan. Several biostimulants were tested (10 in total), including humic acids, macro and micro seaweed extracts, alfalfa protein hydrolysate, amino acids alone or in combination with zinc, B-group vitamins, chitosan and a commercial product containing silicon. Treatments were performed at weekly intervals, starting from the end of May until mid-August. The macroseaweed extract was effective in stimulate tree growth potential in both years, as shown by a significantly larger leaf area (+20% as compared to control) and by an higher chlorophyll content and leaf photosynthetic rate in year 2016. As for the yield performances and apples quality traits at harvest (average fruit weight, soluble solids content, titratable acidity, and flesh firmness), they were generally affected by the different climatic conditions that characterized the two growing seasons (year 2017 being characterized by higher maximal and average temperatures and by limited rainfalls at the beginning of the season). Treatments with macroseaweed extract, B-group vitamins and alfalfa protein hydrolysate were able to significantly improve the intensity and extension of the red coloration of apples at harvest. Correspondingly, the anthocyanin content in the skin of apples treated with the same biostimulants resulted significantly higher than control, highlighting the potential influence of these substances on the synthesis of secondary metabolites in apple. The incidence of physiological disorders was also monitored during apple storage period. Amino acids plus zinc application was effective in reducing (more than 50%) the incidence of the “Jonathan spot,” the main post-harvest disorder for this cultivar.
Yeasts play an important role in the life cycle and biology of the insect pest Drosophila suzukii (Matsumura), commonly known as the spotted wing drosophila (SWD). Adult and larvae of this species are known to feed and benefit from yeast in their diet. In addition, yeasts were found to be attractive to SWD and were repeatedly found to be associated with SWD. Among those, Hanseniaspora uvarum is the most commonly mentioned. The present study explores the chemical composition and the effects of three H. uvarum strains and five yeast species (Saccharomyces cerevisiae, Candida sp., Issatchenkia terricola, Metschnikowia pulcherrima and Saccharomycopsis vini) in the diet of SWD adults. The different yeast species used in this study influenced mortality, fecundity and ingestion by SWD females. Hanseniaspora uvarum and S. vini were preferably ingested and increased fecundity of SWD females. The intra-and extracellular concentrations of compounds, such as amino acids, carbohydrates, sugar alcohols and organic acids, produced or consumed by yeasts differed among the species. Knowledge of the interaction of different yeast species with SWD and specific differences in the profile of compounds of yeast can help to improve the development of control strategies against the insect pest by promoting the ingestion of attractand-kill formulations based on the combinations of yeasts and an appropriate insecticide. Phytoplasmoses such as apple proliferation (AP) and European stone fruit yellows (ESFY) cause severe economic losses in fruit production. A common symptom of both phytoplasma diseases is early yellowing or leaf chlorosis. Even though chlorosis is a well-studied symptom of biotic and abiotic stresses, its biochemical pathways are hardly known. In particular, in this context, a potential role of the senescence-related pheophorbide a oxygenase/phyllobilin (PaO/PB) pathway is elusive, which degrades chlorophyll (Chl) to phyllobilins (PBs), most notably to colorless nonfluorescent Chl catabolites (NCCs). In this work, we identified the Chl catabolites in extracts of healthy senescent apple and apricot leaves. In extracts of apple tree leaves, a total of 12 Chl catabolites were detected, and in extracts of leaves of the apricot tree 16 Chl catabolites were found. The seven major NCC fractions in the leaves of both fruit tree species were identical and displayed known structures. All of the major Chl catabolites were also found in leaf extracts from AP- or ESFY-infected trees, providing the first evidence that the PaO/PB pathway is relevant also for pathogen-induced chlorosis. This work supports the hypothesis that Chl breakdown in senescence and phytoplasma infection proceeds via a common pathway in some members of the Rosaceae family.
The present work was designed to compare the vitamin C (ascorbic acid, AsA) content of pulp, peel, and juice of 64 apple cultivars. These cultivars were carefully identified as 'true to type' by molecular genetic tools, grown in the same site under identical conditions and processed by a standardized protocol. Twenty-one of them, accounting for more than 95% of the apple production of South Tyrol, were chosen to represent the current market, 16 were old or local cultivars formerly grown in the area, and 27 were new cultivars, including 15 with scab resistance and 12 with red flesh fruit. For the determination of the AsA content, a new High-Performance Liquid Chromatography with Diode-Array Detection method was developed and validated. While old cultivars stood out for their high AsA content in pulp and peel, the red-fleshed cultivars are the ones maintaining most of their AsA content during processing. Our data thus suggest a potential for old and red-fleshed cultivars for healthy juices or further processed food components.

In addition, several minor NCC fractions were tentatively classified. The structure of the most polar one of the NCCs, named Pd-NCC-32, featured an unprecedented twofold glycosidation pattern. Three of the NCCs are also functionalized at their 3 2 -position by a glucopyranosyl group. In addition, two of these glycosidated NCCs carry a dihydroxyethyl group at their 18-position. In the polar Pd-NCC-32, the latter group is further glycosidated at the terminal 18 2 -position. Four other major Pd-NCCs and one minor Pd-NCC were identified with five NCCs from higher plants known to belong to the 'epi'-series. In addition, tentative structures were derived for two minor fractions, classified as yellow chlorophyll catabolites, which represented (formal) oxidation products of two of the observed Pd-NCCs. The chlorophyll catabolites in leaves of plum feature the same basic structural pattern as those found in leaves of apple and pear trees.
The production of phagostimulant and attractive volatile organic compounds (VOCs) by yeasts can be exploited to improve the efficacy of attract-and-kill formulations against the spotted wing drosophila (SWD). This study evaluated the persistence over one week of a yeast-based formulation under greenhouse conditions. Potted grape plants were treated with: (i) potato dextrose broth (PDB), (ii) PDB containing spinosad (PDB + S), and (iii) H. uvarum fermentation broth grown on PDB containing spinosad (H. u. + S). Laboratory trials were performed to determine the survival and the oviposition rate of SWD after exposure to treated leaves. Ion-exchange chromatography was performed to measure carbohydrates, sugar alcohols, and organic acids on leaf surfaces, while amino acids were assessed through liquid chromatography–mass-spectrometry. Additionally, the VOCs released by plants treated with H.uvarum were collected via closed-loop-stripping analysis and compared to those emitted by untreated leaves. A higher mortality was observed for adult SWDs in contact with H. uvarum containing spinosad compared to PDB containing spinosad. Generally, a decrease in the amounts of non-volatile compounds was observed over time, though numerous nutrients were still present one week after treatment. The application of the yeast-based formulation induced the emission of VOCs by the treated leaves. The concentration of 2-phenylethanol, one of the main VOCs emitted by yeasts, decreased over time. These findings describe the presence of potential phagostimulants and compounds attractive to SWD in a yeast-based attract-and-kill formulation and demonstrate the efficacy of the formulation over one week.
