Interest in the measurement of salivary cortisol has increased recently because saliva can be easily collected before and after an imposed stress. This study evaluated the relationship between plasma and salivary concentrations of cortisol following ACTH administration in calves (experiment 1) and machine milking of adult cows (experiment 2). A catheter was inserted into the jugular vein of all animals 72 h before the beginning of experiments. Blood and saliva samples were collected before and after ACTH administration (0.6 IU/kg BW) in calves or before and after machine milking of cows. Using a cotton swab, each saliva sample was taken immediately following the blood sample. In general, cortisol profiles were similar in plasma and saliva and correlated in both experiments; however, plasma concentrations were significantly higher than salivary concentrations. In addition, the differences between cortisol concentrations measured in saliva and plasma within each experiment varied substantially between animals and samples. Furthermore, in experiment 2, nearly 10% of salivary samples were below limits of detection. The sharp peaks in cortisol after ACTH administration in both the plasma and saliva were reflected adrenal stimulation. In addition, increases in cortisol in response to milking in both the plasma and saliva suggest that salivary sampling is a reliable option when studying cortisol responses to normal physiological events.

-Milk yield and plasma oxytocin (OT), prolactin (PRL), and cortisol (CORT) during suckling and machine milking were measured in multiparous ewes subjected to a mixed management system of 3 sucklings and two daily milkings. Peak hormones were significantly increased and were similar during suckling and milking for PRL (181 vs. 163.3 ng·mL -1 ) and CORT (12.5 vs. 11.5 ng·mL -1 ). During the period of exclusive suckling, OT was always significantly released (90.3 pg . mL -1 ); however, during the period of mixed management, OT concentrations only increased during suckling compared to milking (91.7 vs. 13.1 pg·mL -1 ). The mean volume of milk obtained during suckling (632 mL) was significantly higher than during milking (255 mL). Thus, during a mixed management system, oxytocin and prolactin releases are not under similar central regulation. A mixed system, without OT release during milking, does not contribute to accelerate the conditioning of ewes for machine milking.

The objective of the present study was to investigate the influence of the establishment of dominance relationships and social stress on plasma cortisol and metabolite levels in Nile tilapia (Oreochromis niloticus). During the 30-day experiment, the fish weighing 236 ± 29 g were kept in individual aquaria, except for two pairings lasting 6 h each. Blood samples were taken from the animals before and after pairing. Display, approach, attack, rebuff, chase flight, and coloration were carried out on days 16 and 30. Activities and behaviors characteristic of the establishment of dominance relationships were described. It was possible to classify all experimental fish (N = 30) as dominant or subordinate. No differences were detected between dominant (N = 15) and subordinate (N = 15) fish during isolation or after pairing in cortisol (isolated: 5.76 ± 0.98 vs 5.42 ± 0.63; paired: 10.94 ± 1.62 vs 11.21 ± 2.45 µg/dl), glucose (isolated: 60.02 ± 4.9 vs 67. 85 ± 16.16; paired: 110.44 ± 15.72 vs 136.26 ± 22.46 mg/dl), triglyceride (isolated: 167.87 ± 5.06 vs 185.68 ± 7.24; paired: 210.85 ± 13.40 vs 221.82 ± 12.70 mg/dl) or total protein levels (isolated: 7.01 ± 0.42 vs 6.69 ± 0.59; paired: 9.21 ± 0.62 vs 9.51 ± 0.66 g/dl). However, when isolated (N = 30) and paired (N = 30) tilapia were compared, there were significant differences in cortisol and metabolite levels. The similar response presented by dominant and subordinate tilapia indicates that establishment of dominance relationships was a stressor for both groups.

RESUMO -Foram estudadas as variações nas concentrações de cortisol e dos hormônios da tireóide de ovinos da raça Corriedale criados em ambiente tropical. Vinte e um ovinos (14 machos e sete fêmeas) foram mantidos em uma câmara climática à 45ºC por nove dias, 8 horas por dia. Registraram-se a temperatura retal (TR) e a frequência respiratória (FR) dos animais, a temperatura ambiente (TA) e a pressão parcial de vapor (PV). Os animais foram posteriormente classificados de acordo com suas respostas quanto à TR e à FR, selecionando-se cinco animais mais aclimatados (grupo A) e cinco animais menos aclimatados (grupo NA) às altas temperaturas do ar. Esses dez animais foram transferidos para um cercado, onde foram registradas a TR e FR duas vezes ao dia e coletadas as amostras sanguíneas uma vez por semana, durante um ano. Foram observadas diferenças nas concentrações de cortisol, de tiroxina (T 4 ) e de triiodotironina (T 3 ), atribuídas às variações na TA e na PV. Ocorreram maiores concentrações de cortisol e menores de T 4 e de T 3 nos períodos de maior TA e/ou PV, sendo que as estimativas dos coeficientes de correlação (r) das variáveis TA e PV com o cortisol foram r = 0,224 e r = 0,395, respectivamente. As correlações entre os hormônios tireoideanos e a TA foram negativas (r = -0,156, para T 4 , e r = -0,151, para T 3 ), evidenciando que um aumento na TA causou redução na taxa metabólica. As variáveis medidas não diferiram entre os grupos de animais classificados. Portanto, a utilização apenas das variáveis fisiológicas TR e FR como parâmetros para a seleção não é suficiente para se avaliar o grau de aclimatação dos animais ao ambiente tropical.Palavras-chave: ambiente tropical, cortisol, hormônios tireoideanos, ovinos Seasonal Variation of Thyroid Hormones and Cortisol of Sheep in Tropical EnvironmentABSTRACT -The variation of cortisol and thyroid hormones in Corriedale sheep bred in a tropical environment was evaluated. Fourteen males and seven females were observed in a climatic chamber at 45ºC for nine days, eight hours per day. Rectal temperature (TR), respiratory rate (FR), air temperature (TA) and air humidity (PV) were recorded. The animals were ranked according to their responses for TR and FR and then the five highest (A group) and five lowest (NA group) ranked animals were selected. These ten animals were moved to a pen where they were observed along one year for TR and FR two times a day, and blood-sampled once a week. The results showed differences for T 3 , T 4 , and cortisol concentrations, which were attributed to variations in TA and PV. Cortisol concentrations were higher and T 3 and T 4 ones were lower under increased TA and PV. Correlations of TA and PV with cortisol values were r = .224 and r = .395 respectively. The negative correlations of TA with T 3 (r = -.151) and T 4 (r = -.156) showed that increase in TA may be followed by decrease in the metabolic rate.
