INTRODUÇÃOOs métodos microbiológicos para avaliação da fertilidade do solo são baseados no principio de que os elementos nutritivos, principalmente nitrogênio, fósforo e potássio que são necessários para o crescimento normal das plantas superiores são exigidos também pelos microorganismos; então, inoculando bactérias e fungos no solo em estudo podemos, através do desenvolvimento conseguido pelos organismos, obter uma informação sobre a reserva do solo em elementos assimiláveis.O uso de A. niger como indicador biológico foi primeiramente sugerido por BUTKEWITSCH (1909) na Rússia. Seu trabalho, entretanto, não recebeu atenção até 1928 quando BE-NECKE e SODING usaram o princípio para determinar deficiências de fósforo e potássio em solos comparando o crescimento do micélio em meio nutritivo líquido contendo quantidades conhecidas de fósforo e potássio com o crescimento verificado em meio nutritivo ao qual haviam sido adicionadas pequenas quantidades de solo. O método foi mais tarde desenvolvido em bases quantitativas por NIKLAS e colaboradores (1930, a, b, 1932, 1936) para P, K e Mg: foi verificada boa concordância 'entre os rseultados do método de A. niger e o de NEUBAÜER-SCHNEIDER.O método de NIKLAS foi modificado por MEHLICH et al. (1933). No ano seguinte, MEHLICH et al. (1934) apresentaram o método da placa de Cunninghamella aplicável a P somente; os diâmetros das colônias de Cunninghamella crescendo em placas contendo quantidades crescentes de P seguem a equação de MITSCHERLICH por interpolação na qual podemos determinar a quantidade de P assimilável que o solo possui. Em 1938Em , 1939, MULDER modificou a técnica de NIKLAS para fazer determinações quantitativas de cobre e magnésio assimiláveis dos solos.(Para uma revisão mais detalhada da literatura, ver VAN-DECAVEYE, 1948, pp. 215-227 0,0007 g Água destilada 1.000 ml 40 ml. de solução nutritiva eram adicionados a uma série de balões de Erlerimeyer de 125 ml. O enxofre era adicionado como Na2SO4.10 H20 nas seguintes proporções : nihil, 100, 200, 300, 400 500, 600, 800, 1000, 1500 e 2000 y calculados como S elementar. Amostras em duplicata de 2 solos bem diferentes, uma terra roxa de bôa fertilidade e um solo arenoso pobre'eram adcionadas ao meio nutritivo sem enxofre. As soluções nutritivas eram esterilizadas em autoclave a 105°C. durante 30 minutos. Então, a solução em cada frasco era inoculada com 0,5 ml de uma densa suspensão de esporos preparada lavando o crescimento de 5 dias de idade de 4 tubos de cultura de A. niger (linhagem isolada na E. S. A. "Luiz de Queiroz", Piracicaba, S. Paulo, Brasil). As culturas eram incubadas a 35°C. durante 5 dias. No fim desse período o micélio era colhido, lavado com água distilada e seco primeiro a 70-90°C. durante 12 horas e depois a 90-100°C. por uma hr.
