BackgroundPreadipocyte differentiation plays a critical role in subcutaneous fat deposition in pigs. However, the roles of different RNAs, such as messenger RNAs (mRNAs), long non-coding RNAs (lncRNAs), and circular RNAs (circRNAs) in the differentiation process of subcutaneous preadipocytes, are still largely unclear. In the present study, a transcriptome analysis, including the analysis of mRNAs, lncRNAs, and circRNAs, during different differentiation stages, namely, day 0 (D0), day 2 (D2), day 4 (D4), and day 8 (D8), of subcutaneous preadipocytes from Chinese Erhualian pigs was performed.ResultsA total of 1554, 470, 1344, 1777, and 676 differentially expressed (DE) mRNAs, 112, 58, 95, 136, and 93 DE lncRNAs, and 902, 787, 710, 932, and 850 DE circRNAs were identified between D2 and D0, between D4 and D2, between D8 and D4, between D4 and D0, and between D8 and D0, respectively. Furthermore, functional enrichment analysis showed that the common DE mRNAs during the entire differentiation process were mainly involved in lipid metabolic and cell differentiation processes. Additionally, co-expression network analysis identified the potential lncRNAs related to adipogenesis, e.g., MSTRG.131380 and MSTRG.62128.ConclusionsOur study provides new insights of the expression changes of RNAs during adipogenic differentiation, which might contribute to the phenotype of subcutaneous adipogenesis. Previous analyses of plant growth-promoting bacteria (PGPB) combined with the remediation of heavy metal pollution in soil have largely been performed under potting or greenhouse conditions, and in situ remediation experiments under field conditions have rarely been reported. In this study, the effects of the metal-resistant PGPB Microbacterium oxydans JYC17, Pseudomonas thivervalensis Y1-3-9, and Burkholderia cepacia J62 on soil Cu pollution under rape remediation were studied in the farmland surrounding the Nanjing Jiuhuashan copper mining region in China. Following inoculation treatment for 50 days, the biomasses of the rape inoculated with strains JYC17, Y1-3-9, and J62 increased, and the total amounts of Cu uptake increased by 113.38, 66.26, and 67.91%, respectively, the translocation factor (TF) of rape inoculated with J62 was 0.85, a significant increase of 70.68%, thus improving the Cu remediation efficiency of the rape. Y1-3-9 and J62 affected the bioavailability of Cu in the soil, and the water-soluble Cu contents were increased by 10.13 and 41.77%, respectively, compared with the control. The antioxidant activities in the rape leaves showed that the tested bacteria increased the contents of antioxidant non-enzymatic substances, including ascorbic acid (ASA) and glutathione (GSH), which were increased by 40.24–91.22% and 9.89–17.67%, respectively, thereby reducing the oxidative stress caused by heavy metals and the contents of thiobarbituric acid-reactive substances (TBARS) and peroxidase (POD). PCR-denaturing gradient gel electrophoresis (PCR-DGGE) was used to analyze the effects of the tested bacteria on the cultivation-dependent and cultivation-independent bacterial communities in the root endosphere and rhizosphere soil of the rape. The sequencing results of the DGGE bands indicated that the tested bacteria colonized the endosphere and rhizosphere, and they became an important component of the cultivation-dependent bacteria. The canonical correspondence analysis (CCA) of the DGGE profile and similarity cluster analysis showed that the tested bacteria affected the cultivation-dependent and cultivation-independent bacterial communities in the root endosphere and rhizosphere. In this experiment, the effects and mechanisms of the combined plant-microbe remediation under field conditions were preliminarily studied, and the results are expected to provide a theoretical basis for future combined remediation experiments.

The coprecipitation of heavy metals (HMs) with Fe­(III) in the presence of dissolved organic matter (DOM) is a crucial process to control the mobility of HMs in the environment, but its underlying immobilization mechanisms are unclear. In this study, Cr­(III) immobilization by coprecipitation with Fe­(III) in the presence of straw-derived DOMs under different Fe/C molar ratios, pHs, and ionic strengths was investigated using scanning transmission X-ray microscopy (STXM) and ptychography and X-ray absorption near-edge structure (XANES) spectroscopy. The results showed that Cr­(III) retention was enhanced in the presence of DOM, a maximum of which was achieved at an Fe/C molar ratio of 0.5. The increase of pH and ionic strength could also promote Cr­(III) immobilization. Cr K-edge XANES results indicated that Fe (oxy)­hydroxide fractions, instead of organics, provided the predominant binding sites for Cr­(III), which was directly confirmed by high spatial resolution STXM-ptychography analysis at the sub-micron- and nanoscales. Moreover, organics could indirectly facilitate Cr immobilization by improving the aggregation and deposition of coprecipitate particles through DOM bridging or electrostatic interactions. Additionally, C K-edge XANES analysis further indicated that the carboxylic groups of DOM were complexed with Fe (oxy)­hydroxides, which probably contributed to DOM bridging. This study provides a new insight into Cr­(III) immobilization mechanisms in its coprecipitation with Fe­(III) and DOM, which could have important implications on the management of Cr­(III)-enriched soils, particularly with crop straw returning.
