SummaryDespite the ubiquity and beneficial role of silicon (Si) in plant biology, structural and chemical mechanisms operating at the single-cell level have not been extensively studied.To obtain insights regarding the effect of Si on individual cells, we cultivated suspended rice (Oryza sativa) cells in the absence and presence of Si and analyzed single cells using a combination of physical techniques including atomic force microscopy (AFM).Si is naturally present as a constituent of the cell walls, where it is firmly bound to the cell wall matrix rather than occurring within intra-or extracellular silica deposition, as determined by using inductively coupled plasma mass spectrometry (ICP-MS) and X-ray photoelectron spectroscopy (XPS). This species of Si, linked with the cell wall matrix, improves the structural stability of cell walls during their expansion and subsequent cell division. Maintaining cell shape is thereby enhanced, which may be crucial for the function and survival of cells.This study provides further evidence that organosilicon is present in plant cell walls, which broadens our understanding of the chemical nature of 'anomalous Si' in plant biology.

Although plant nutriomics is still at a conceptual stage, substantial efforts are being made in China aimed at increasing plant nutrient efficiency through a nationwide, co-ordinated project on plant nutriomics. Future studies involving both national and international collaborations are needed to develop nutrient-efficient, stress-tolerant and high-quality crop varieties for both China and elsewhere.

Aquaporins (AQPs) are an abundant protein family and play important roles to facilitate small neutral molecule transport across membranes. Oilseed rape (Brassica napus L.) is an important oil crop in China and elsewhere in the world, and is very sensitive to low boron (B) stress. Several AQP family genes have been reported to be involved in B transport across plasma membranes in plants. In this study, a total of 121 full-length AQPs were identified and characterized in B. napus (AC genome), and could be classified into four sub-families, including 43 PIPs (plasma membrane intrinsic proteins), 35 TIPs (tonoplast intrinsic proteins), 32 NIPs (NOD26-like intrinsic proteins), and 11 SIPs (small basic intrinsic proteins). The gene characteristics of BnaAQPs were similar to those of BraAQPs (A genome) and BolAQPs (C genome) including the composition of each sub-family, gene structure, and substrate selectivity filters. The BnaNIP was the most complex AQP sub-family, reflecting the composition of substrate selectivity filter structures which affect the permeation of solution molecules. In this study, the seedlings of both B-efficient (QY10) and B-inefficient (W10) cultivars were treated with two boron (B) levels: deficient (0.25 μM B) and sufficient (25 μM B). The transcription of AQP genes in root (R), juvenile leaf (JL), and old leaf (OL) tissues of both cultivars was investigated under B deficient and sufficient conditions. Transcription of most BnaPIPs and BnaTIPs was significantly increased compared with other BnaAQPs in all the three tissues, especially in the roots, of both B-efficient and B-inefficient cultivars under both B conditions. With B deprivation, the expression of the majority of the BnaPIPs and BnaTIPs was down-regulated in the roots. However, the BnaNIPs were up-regulated. In addition, the BnaCnn_random.PIP1;4b, BnaPIP2;4s, BnaC04.TIP4;1a, BnaAnn_random.TIP1;1b, and BnaNIP5;1s (except for BnaA07.NIP5;1c and BnaC06.NIP5;1c) exhibited obvious differences at low B between B-efficient and B-inefficient cultivars. These results will help us to understand boron homeostasis in B. napus.

Boron (B) is an essential micronutrient for plants, but the molecular mechanisms underlying the uptake and distribution of B in allotetraploid rapeseed (Brassica napus) are unclear. Here, we identified a B transporter of rapeseed, BnaC4.BOR1;1c, which is expressed in shoot nodes and involved in distributing B to the reproductive organs. Transgenic Arabidopsis plants containing a BnaC4.BOR1;1c promoter-driven GUS reporter gene showed strong GUS activity in roots, nodal regions of the shoots and immature floral buds. Overexpressing BnaC4.BOR1;1c in Arabidopsis wild type or in bor1-1 mutants promoted wild-type growth and rescued the bor1-1 mutant phenotype. Conversely, knockdown of BnaC4.BOR1;1c in a B-efficient rapeseed line reduced B accumulation in flower organs, eventually resulting in severe sterility and seed yield loss. BnaC4.BOR1;1c RNAi plants exhibited large amounts of disintegrated stigma papilla cells with thickened cell walls accompanied by abnormal proliferation of lignification under low-B conditions, indicating that the sterility may be a result of altered cell wall properties in flower organs. Taken together, our results demonstrate that BnaC4.BOR1;1c is a AtBOR1-homologous B transporter gene expressing in both roots and shoot nodes that is essential for the developing inflorescence tissues, which highlights its diverse functions in allotetraploid rapeseed compared with diploid model plant Arabidopsis.

The importance of SPX domain-encoding proteins to phosphate (Pi) homeostasis and signaling pathways has been well-documented in rice and Arabidopsis. However, global information and responses of SPX members to P stress in allotetraploid Brassica napus, one of the world’s major oil crops that is sensitive to P deficiency, remain undefined. We identified a total of 69 SPX domain-containing genes in the B. napus genome. Based on the domain organizations, these genes were classified into four distinct subfamilies—SPX (11), SPX-EXS (43), SPX-MFS (8), and SPX-RING (7)—that represented clear orthologous relationships to their family members in Arabidopsis. A cis-element analysis indicated that 2 ∼ 4 P1BS elements were enriched in the promoter of SPX subfamily genes except BnaSPX4s. RNA-Seq analysis showed that BnaSPX genes were differentially expressed in response to Pi deficiency. Furthermore, quantitative real-time reverse transcription PCR revealed that nine SPX subfamily genes were significantly induced by Pi starvation and recovered rapidly after Pi refeeding. A functional analysis of two paralogous BnaSPX1 genes in transgenic Arabidopsis indicated their functional divergence during long-term evolution. This comprehensive study on the abundance, molecular characterization and responses to Pi deficiency of BnaSPX genes provides insights into the structural and functional diversities of these family members in B. napus and provides a solid foundation for future functional studies of BnaSPX genes.Highlight: The genome-wide identification and characterization of SPX genes in B. napus and their responses to Pi deficiency provide comprehensive insights into the structural and functional diversities of the family members in B. napus and their potential in Pi homeostasis and signaling responsiveness to Pi stress.
