FOXO1 protein inactivation in the nucleus is one of targets for the treatment of diabetes mellitus. Annona muricata leaves contain flavonoid and phenolic compound alkaloids that were known to be able to increase pancreatic b cell proliferation in animal experiment. This research aimed to predict the active compound ability of the Annona muricata leaves to bind and inhibit FOXO1 protein through in silico study. Analysis of molecular docking was performed by using Autodock Vina PyRx. this research proved that anonaine, rutin, muricatocin a, isolaureline, xylopine, and kaempferol 3-O-rutinoside had an equal or smaller free binding energy compared to the control compound. Rutin and Muricatocin A had the same binding ability toward 66% amino acid residues, compared to control compound with hydrogen bond type, while xylopine, anonaine, isolaureline, kaempferol 3-O-rutinoside had a similar binding ability towards 33% amino acid residues compared to control compound with hydrogen bond type.
Inflammatory reactions give rise to a series of processes and symptoms that can cause pain and damage to nearby cells. Inflammatory reactions are mediated by inflammatory and proinflammatory cytokines. NSAID class (non-steroidal anti-inflammatory drug) drugs have been developed to inhibit the inflammatory process by suppressing COX-1 and / or COX-2 enzyme activity. However, this drug can cause gastric mucosal irritation. the ethanol extract of Annona muricata leaf has an analgesic and anti-inflammatory effect[3].The purpose of this research is to know analgesic and antiinflamatory activity from ethanol extract and essential oil of Annona muricata leaf.The method used was the study of in silico by molecular docking of active compounds in ethanol extracts and essential oils to the COX-1, COX-2, phospolipase-A, and TNF-alpha receptor molecules as target molecules. The molecular docking results show that Annona muricata ethanol extract can act as an anti-inflammatory and analgesic, especially through strong bonds between "rutine" compounds with COX-2 receptors with affinity -10 kcal / mold and "quercetin" with affinity bindings -9.8 kkal.mol, better than dexametason (binding affinity -9.6 kcal / mol). Objective: Soursop leaf contains alkaloids, terpenoids, flavonoids, acetogenin, and phenolics. Soursop leaf decoction is usually consumed as an anti-obesity agent, even though its active compounds and its action mechanism are still unclear. Computational approaches have been developed to predict the ability of an active compound to bring about biological effects. This study was designed to predict the potency of soursop (Annona muricata Linn.) water extract for the activating of Glucagon-Like Peptide-1 Receptor (GLP-1R), Inhibiting Dipeptidyl Peptidase 4 (DPP4), and Forkhead Box Protein O1 (FoxO1) protein using in silico analysis. Methods: Identification of active compounds contained in soursop leaf water extract was carried out using physicochemical methods and Gas chromatography–mass spectrometry (GC-MS). They were then analyzed using computational analysis, i.e., potential analysis using the Way2drug web server. Protein interaction predictions of GLP-1R with the active compounds found in soursop leaf water extract using STITCH. The affinities of the active compounds of soursop leaf to the proteins DPP4 and FoxO1 were also analyzed using molecular docking. Results: Active compounds of the soursop leaf water extract contain 5-isopropenyl-3,8-dimethyl-1,2,3,3A, 4,5,6,7-octa hydro azulene (22.17%) and 1,2-benzene dicarboxylic acid, diethyl ester (phthalic acid) (57.30%). The active ingredients have not been shown to interact with GLP-1R. 5-isopropenyl-3,8-dimethyl-1,2,3,3A, 4,5,6,7-octa hydro azulene and phthalic acid both have a weak affinity for DPP4, and only phthalic acid has a weak affinity with the FoxO1 protein. Conclusion: Phthalic acid has a weak potential as an inhibitor of the DDP4 and FoxO1 proteins.
Desa Sukodono Kecamatan Dampit Kabupaten Malang merupakan salah satu daerah tujuan KKN Universitas Islam pada periode Pebruari 2020. Tujuan Kegiatan pengabdian kepada masyarakat (PPM) dalam rangka kegiatan KKN yang dilakukan oleh kelompok 63 adalah (1) Meningkatkan perekonomian masyarakat di Desa Sukodono dengan cara pembuatan produk olahan selai kopi, (2) Meningkatkan movitasi belajar siswa di SDN 1 Sukodono, dan (3) Meningkatkan tindakan preventif dan promotif stunting di Desa Sukodono. Kegiatan dilakukan melalui metode penyuluhan dan pelatihan, diskusi dan tanya jawab, serta demonstrasi secara langsung. Hasil akhir dari kegiatan menunjukkan bahwa kegiatan pengabdian kepada masyarakat (PPM) mampu memberikan motivasi dan perubahan pola fikir kepada masyarakat di Desa Sukodono terutama tentang bagaimana cara membuat produk olahan dari serbuk kopi, meningkatkan motivasi belajar siswa dan meningkatkan tindakan preventif dan promotif stunting. Kendala-kendala yang muncul dapat diatasi dengan metode pelatihan secara bertahap. Adanya MOU kerjasama dengan pemerintah daerah dan pemerintah desa perlu dilakukan untuk keberlajutan program secara kontinue dan dievaluasi keberhasilannya dalam meningkatkan ekonomi, kesejahteraan dan penurunan angka stunting di Kabupaten Malang.
Introduction: Rifampicin is one of anti-tuberculosis drugs are reported to have hepatotoxic effects through increasing ROS (Reactive Oxygen Species) so thattriggering necrosis of liver cells and stimulates the formation of TNF-α. Essential oil of Annona muricata Linn. contains linalool, β-caryophyllen, trans-caryophyllen, germacrene A, germacrene B, germacrene D, sesquisterpene, β-canidine, γ-canidine. The objective of this study is to investigate the effect of essential oil of Annona muricata Linn. on levels of Tumor Necrosis Alpha(TNF-α) liver in male wistar rats which induced by rifampicin.Methods: This was an invivo experimental study with control group post test only design and use 25 male wistar rats and divided into 5 groups, they are KN (corn oil), KP (Rifampicin), P1 (Rifampicin + Essential oil 1,5%), P2 (Rifampisin + Essential oil 3%), P3 (Rifampisin + Essential oil 6%) for 6 weeks and then levels of TNF-α liver will be checked. Analysis of the data using One Way (ANOVA) test and then tested by post hoc test using Least Significant Difference (LSD). The result significant if p<0,05.Result: Induced rifampicin can increased levels of TNF-α liver than group KN (p<0,05). Treatment of essential oil of Annona muricata Linn. on P1, P2, and P3 can reduced levels of TNF-α liver than group KP (p<0,05). Allegedly this is due to the active ingredient in the essential oils act as antioxidants and antiinflammatory.Conclusion: Essential oil of Annona muricata Linn. at all doses can reduced levels of TNF-α liver in male wistar rats which induced by rifampicin.Keywords:rifampicin,essential oil of Annona muricata Linn., TNF-α liver
