A eficiência dos procedimentos de digestão em fornos de microondas pode ser afetada por parâmetros experimentais críticos, como massa de amostra, volume da mistura ácida, potência de radiação aplicada e o tempo da digestão. Neste trabalho foi proposta a utilização de um planejamento fatorial completo para a otimização da digestão de amostras de óleo lubrificante em forno de microondas, com recipientes fechados. O teor de carbono residual e a acidez final das soluções foram determinados em todos os digeridos provenientes do forno de microondas, como resposta ao planejamento fatorial. Cálcio, Cu, Mg, P, S e Zn foram determinados nos digeridos ácidos e a exatidão foi checada utilizando-se um material de referência certificado, NIST SRM 1848-Lubricating Oil Additive Package. Os resultados encontrados para as amostras e para o material de referência certificado estão em concordância a um nível de confiança de 95%. Tempo de digestão, potência de radiação aplicada, massa de amostra e as interações entre essas variáveis foram significativas (P < 5%), aplicando-se a análise de variância (ANOVA).This work proposes the use of factorial design for optimization of microwave-assisted digestion of lubricating oil. The accuracy of digestion procedures is affected by critical experimental parameters, such as sample amount, concentrated acid volumes, microwave radiation applied power, and digestion time. The effects of these key variables on the microwave-assisted digestion efficiency were investigated. The residual carbon content and the acidity were determined in all digestates after microwave-assisted digestion as response of the factorial design. Calcium, Cu, Mg, P, S, and Zn were determined in oil digestates obtained by using two systems: a cavity-and a focused-oven. The accuracy was checked using one standard reference material, NIST SRM 1848 -Lubricating Oil Additive Package. All determined and certified values are in agreement at a 95% confidence level. A decomposição de amostras que contêm elevados teores de Si, utilizando mistura com os ácidos nítrico e fluorídrico, pode provocar a precipitação de fluoretos de Al(III), Ca(II), Fe(III) e Mg(II) durante a digestão, impedindo a recuperação total desses elementos. Visando minimizar esse problema, foram testados quatro procedimentos aplicados a diferentes amostras de solos. O procedimento mais eficiente envolveu a decomposição assistida por radiação microondas, seguida pela separação do material sólido residual com elevado teor de Si do sobrenadante ácido. A fração residual foi solubilizada usando HF concentrado e, após complexação com ácido bórico, essa solução foi misturada à fase líquida do digerido. Os teores de metais nos digeridos foram determinados por espectrometria de emissão óptica em plasma induzido (ICP OES) e a exatidão foi demonstrada aplicando o procedimento proposto em amostras provenientes do programa colaborativo International Soil-Analytical Exchange (ISE, Wegening Agricultural University, Holanda) e em amostra certificada de solo proveniente do National Institute of Standard and Technology (NIST, SRM 2709), que apresentou respectivamente, os seguintes percentuais de recuperação: 100,0 ± 2,6; 99,5 ± 1,9; 108,5 ± 2,7; 94,6 ± 8,9; 92,2 ± 9,7; 95,7 ± 1,8; 96,7 ± 2,7; 95,2 ± 0,6; 103,6 ± 2,6 e 96,1 ± 1,6 para, Al, Ba, Ca, Cr, Cu, Fe, Mg, Mn, V e Zn.Termos de indexação: digestão de solo, ácido fluorídrico, análises de solo, solos.(1) Recebido para publicação em agosto de 2004 e aprovado em maio de 2005.

An analytical method for the isolation based on matrix solid-phase dispersion technique and gas chromatographic determination of pesticides in cattle plasma is presented. It was fortified 0.25 g of plasma with pesticides and blended with 1 g each C18 and Na 2 SO 4 . The homogenized matter was transferred to a SPE cartridge, which contained 1 g of activated florisil with 5 mL acetonitrile. The analites were eluted under vaccum with 15 mL acetonitrile, the extract was analyzed by gas chromatography-mass spectrometry. The limit of quantification of the method was 0.04 mg L -1 for chlorphenvinfos and fipronil and 0.02 mg L -1 for cypermethrin..Keywords: pesticides; MSPD; GC-MS. INTRODUÇÃOOs medicamentos veterinários são empregados na medicina veterinária para fins terapêuticos, profiláticos ou como promotores de crescimento. Seu uso inadequado, pela não observância do tempo de carência, dosagens incorretas, via de administração não recomendada, terapia indiscriminada e uso de substâncias proibidas podem deixar resíduos em produtos alimentícios de origem animal, colocando em risco a saúde humana.1 No Brasil 57% do mercado veterinário é destinado a bovinos, destacando os antimicrobianos, endoparasitas, ectoparasitas, promotores de crescimento e antibióticos. 2O Brasil detém o segundo maior rebanho bovino comercial do mundo, sendo o maior exportador mundial de carne, 3 mas apresenta deficiências no controle de qualidade, tanto microbiológica, como também relacionado à presença de resíduos de substâncias químicas nos produtos de origem animal disponível no mercado. 4 A presença de resíduos nos alimentos, destacando os pesticidas que são aplicados no rebanho bovino, no combate ao carrapato (Boophilus microplus) e moscados-chifres (Haematobia irritanis), podem deixar resíduos no leite e na carne comercializada, comprometendo a saúde da população se estiverem em níveis acima dos permitidos pela legislação (Food and Agriculture Organization -FAO e Organização Mundial da Saúde -OMS), podendo inibir as relações de exportação/importação entre países. 5Os métodos analíticos para monitorar resíduos de medicamentos veterinários em matrizes alimentícias, destacando os pesticidas que são utilizados no combate a ectoparasitas do rebanho bovino, devem ser rápidos, específicos e sensíveis, para permitir a detecção a baixas concentrações.As análises de amostras de alimentos sólidas ou semissólidas, destacando as de origem biológica, são extremamente trabalhosas.
