Differential Adjuvant Effects of Soluble Beta Glucans from Barley and Saccharomyches Cerevisia in Primary and Secondary Humoral Immune Responses

Abstract

The purpose of this study was to investigate and compare adjuvant effects of soluble ß-glucans from barley and Saccharomyches cerevisia in induction of antigen specific humoral immune responses. Mice were immunized with conalbumin at a relatively low concentration in the presence of beta glucans. Anti-conalbumin antibodies in the sera of immunized and control mice were quantified by ELISA. At high doses, both of glucans increased effectively levels of circulating IgM and IgG antibodies which were specific for conalbumin. However, coadministration of glucan from barley at 1 µg dose resulted in lower yield in IgG1, IgG2a, IgG2b, and IgA levels in comparison to that of yeast-derived glucan at the same dose. We also found that antigen (conalbumin) specific antibody levels enhanced by ß-glucan from Saccharomyches cerevisia were always higher than those of the glucan from barley. Based on these findings, we concluded that ( ÖZET Saccharomyches cerevisia ve Arpadan Elde Edilen Beta Glukanlar›n Primer ve Sekonder Hümoral ‹mmün Cevaptaki Farkl› Adjuvan EtkisiBu çal›flman›n amac› antijene özgül hümoral immün cevab›n oluflmas› s›ras›nda arpa ve Saccharomyches cerevisia'dan elde edilen çözünebilir ß-glukanlar›n adjuvan etkilerinin araflt›r›lmas›d›r. Fareler beta glukan varl›¤›nda göreceli olarak düflük dozlarda konalbümin ile immünize edildi. ‹mmünize edilmifl ve edilmemifl kontrol fare serumunda anti-konalbumin antikorlar› ELISA yöntemi ile tayin edildi. Yüksek dozlarda uyguland›¤› zaman her iki glukan konalbümine özgül IgM ve IgG antikor seviyelerini etkin bir flekilde artt›rd›. Ancak, ayn› dozda maya (Saccharomyches cerevisia) kökenli glukan ile karfl›laflt›r›ld›¤›nda, konalbümin ve 1 µg arpadan elde edilen glukan›n birlikte enjeksiyonu, daha düflük seviyelerde IgG1, IgG2a, IgG2b, and IgA üretimine neden oldu. Saccharomyches cerevisia köken-li glukan taraf›ndan artt›r›lan antijene özgül antikor seviyeleri, arpa kökenli glukan taraf›ndan artt›r›lan antijene özgül antikor seviyelerinden daima daha fazla oldu. Bu sonuçlar temel al›narak antijene özgül hümoral immün yan›t oluflmas›nda, mayadan elde edilen (1,3),( 1,6)-ß-D-glukan›n, arpadan elde edilen (1,3),( 1,4)-b-D-glukan'a göre daha fazla immünostimülan aktiviteye sahip oldu¤u düflünüle-bilir.
