Zika virus infection leads to mitochondrial failure, oxidative stress and DNA damage in human iPSC-derived astrocytes

Abstract: Zika virus (ZiKV) has been extensively studied since it was linked to congenital malformations, and recent research has revealed that astrocytes are targets of ZiKV. However, the consequences of ZiKV infection, especially to this cell type, remain largely unknown, particularly considering integrative studies aiming to understand the crosstalk among key cellular mechanisms and fates involved in the neurotoxicity of the virus. Here, the consequences of ZiKV infection in ipSc-derived astrocytes are presented. our… Show more

“…Once the integrity of BBB, which is composed of specialized microvascular endothelial cells, is crossed, the virus is able to reach and infect its target cells, and thus, presentation of rare clinical events may be observed [107]. Indeed, a number of studies have shown the prominent role of astrocytes in ZIKV infection [108][109][110][111][112], which may play a role in ZIKV infection in the brain.…”

“…This dysregulation has been previously observed during placental transcriptome profiling 29 and may be related to an overall cellular energy imbalance induced by ZIKV infection. 54 In summary, we conclude that ZIKV differs from other flaviviruses in downregulating IGF2 gene expression, although further studies are necessary to investigate the mechanisms involved. We also showed that mutations in the ZIKV genome may result in differences in the biology of the viruses and, consequently, the host response.…”

“…Accordingly, it has been speculated that blood pressure elevation might result in development of hypertension [23]. Furthermore, it is known that DNA undergoes oxidative damage from oxidant stress [24,25], while others have shown that tetradecanoyl phorbol (TPA), a carcinogen formed by DNA damage, induces activation of breast cancer cells [26][27][28]. Therefore, it is likely that an increased concentration of active oxygen might impair mammary gland cells, leading to onset of mammary gland disease.…”
