Wnt1 Accelerates anEx VivoExpansion of Human Cord Blood CD34+CD38Cells

Abstract: Cord blood hematopoietic stem cells (CB-HSCs) transplantation has been increasing gradually with facing the limitation of insufficient quantity of HSCs in each CB unit. Therefore, efficient expansion methods which can maintain stem cell characteristics are needed. In this study, umbilical CB-CD34+ cells were cultured in two different cytokine cocktails: 4 factors (4F = Flt3-L, SCF, IL-6, and TPO) and 5 factors (5F = Wnt1 + 4F) in both serum and serum-free media. The data revealed that the best condition to acc… Show more

“…Neutrophils are essential for the innate immune response and can protect patients from bacterial infections in the initial phase of transplantation, especially for patients undergoing myelosuppressive chemotherapy [38,39]. Therefore, the capability to generate matured blood cells is also critical for stem cell transplantation [11,40].…”

“…6). To compensate for the limitation of AKTi in human HSPC expansion, other factors, including Wnt activator [38,41,42], should be used in combination with AKTi. We expect that the combination of AKTi and the activator of canonical Wnt signaling will promote the ex vivo expansion of functional HSPCs without affecting their differentiation capability (Fig.…”

“…Recently, it has been demonstrated that the miRNA-320 family suppresses stem cell-like characteristics in prostate cancer cells by down-regulating the Wnt/beta-catenin signaling pathway [ 49 ]—a pathway which is also of central importance for regulation of self-renewal and proliferation in hematopoietic stem cells [ 50 , 51 ]. Furthermore, miRNA-320 targets the transferrin receptor CD71 in the human leukemia cell line HL-60, thereby inhibiting proliferation [ 52 ].…”
