Volvopluteus and Pluteus section Pluteus (Agaricales: Pluteaceae) in Turkey based on morphological and molecular data

Abstract: Pluteus and Volvopluteus are two closely related genera of Pluteaceae that comprise saprotrophic fungi. Although the genera have been well-studied in Europe, North and South America, the diversity of Pluteus, mainly Pluteus sect. Pluteus, and Volvopluteus in Turkey is poorly known due to insufficient morphological descriptions and a lack of DNA sequence data. In this study, the diversity of Pluteus sect. Pluteus and Volvopluteus in Turkey is evaluated by employing both morphological features and phylogenetic a… Show more

“…The type material of P. pallescens , deposited in K, was studied by B. Dima, and the molecular methods followed [ 65 , 66 ]. The total genomic DNA of OK collections was extracted in accordance with Kaygusuz et al [ 67 ]. For collections in LE, the DNA extraction procedure completely corresponded to the manufacturing protocol of the Phytosorb Kit (ZAO Syntol).…”

“…The genomic DNA was extracted from dried specimens using ZR Fungal/Bacterial DNA MiniPrep kit (Zymo research, Irvine, California) according to the manufacturer's protocol. Polymerase chain reaction was performed as described by Kaygusuz et al (2021) using the primer pairs ITS1F/ITS4 (White et al, 1990) and LR0R/LR5 (Vilgalys and Hester, 1990) to amplify nrITS and nLSU gene regions, respectively. Same primer sets were used for sequencing reactions (Source Bioscience, Berlin, Germany).…”

“…Microscopic structures were observed in dried material stained in ammoniacal Congo red. The following abbreviations are used: L m and W m indicate the average length and width of basidiospores, Q shows the ratios of length/width and Q m presents the average quotient of the measured basidiospores [5]. Specimens are kept at the fungarium of Isparta University of Applied Sciences, Isparta, Turkey.…”

“…DNA isolation, polymerase chain reaction (PCR) amplifications and sequencing methods were carried out according to Kaygusuz et al [5]. The primer pairs ITS1F/ITS4 [6,7] and LR0R/LR5 [8] were used for amplifying nrITS and nrLSU gene regions, respectively.…”
