Salinity stress is one of the most common abiotic stresses that hamper plant productivity worldwide. Successful plant adaptations to salt stress require substantial changes in cellular protein expression. In this work, we present a 2-DE-based proteomic analysis of a model unicellular green alga, Chlamydomonas reinhardtii, subjected to 300 mM NaCl for 2 h. Results showed that, in addition to the protein spots that showed partial up- or down-regulation patterns, a number of proteins were exclusively present in the proteome of the control cells, but were absent from the salinity-stressed samples. Conversely, a large number of proteins exclusively appeared in the proteome of the salinity-stressed samples. Of those exclusive proteins, we could successfully identify, via LC-MS/MS, 18 spots uniquely present in the control cells and 99 spots specific to NaCl-treated cells. Interestingly, among the salt-exclusive protein spots, we identified several important housekeeping proteins like molecular chaperones and proteins of the translation machinery, suggesting that they may originate from post-translational modifications rather than from de novo biosynthesis. The possible role and the salt-specific modification of these proteins by salinity stress are discussed.

Three Allium vegetables, Welsh onion from Okinawa, and Wakegi from Okinawa and Nagasaki were subjected to salinity-stress using seawater. The K + /Na + ratio of the three Allium vegetables clearly decreased; however, there was no significant effect on growth parameters. Of the three Allium vegetables, 10% seawater treatment of Welsh onion and Wakegi from Nagasaki clearly enhanced both the sugar content and antioxidant activity. Therefore, seawater treatment may be potentially useful for the development of value-added Allium vegetables, enhancing the palatability and food functionality.Keywords: Allium vegetable, salinity-stress, seawater, sugar content, antioxidant activity IntroductionSalinity is a major environmental factor limiting crop growth and productivity (Rhodales and Loveday, 1990). During the onset and development of salinity-stress within a crop plant, major processes such as photosynthesis, protein synthesis, and energy and lipid metabolism are affected, leading to qualitative and yield losses in most crops (Hasegawa et al., 2000; Hagemann and Erdmann, 1997; Hayashi and Murata 1998). However, there are several crops with an inherent capacity to withstand salinity-stress, which allows for stable vegetable production and significantly contributes to palatability and food functionality (Sato et al., 2006). Thus, there have been several reports on the application of salinity-stress for improving the quality of vegetables such as tomato fruits (Auerswald et al., 1999), spinach (Makabe and Tanii, 2008), and strawberry (Keutgen and Pawelzik, 2000).Welsh onion (Allium fistulosum L.), a member of the Allium family, is a popular vegetable in Asian countries and a reputed good source of phenol compounds with antioxidant activity, leading to beneficial medicinal effects (Helen et al., 2000;Wang et al., 2006).Hence, we focused our attention on exploiting salinity-stress treatment for the development of value-added Welsh onion with augmented palatability and food functionality. There are relatively few studies on the salt tolerance of Welsh onion. In this study, we investigate the effects of artificial salinity-stress on the growth, relative chlorophyll content, and K + /Na + ratio of Allium vegetables: Welsh onion and Wakegi (Allium x wakegi A.), and assess the effect of salinity-stress on sugar content, total phenol content, and antioxidant activity. Chloride and carbonate salts are the main salts causing salinization and widely exist in aquatic environment, so algae may suffer from salinization stress for high water evaporation. In this study, in order to investigate and compare the toxic effects of the two salts on algal photosynthesis, we used NaCl and Na2CO3 to stress Chlamydomonas reinhardtii. Under the two salt stresses, the content of O −.2 and H2O2 in the cells was increased significantly, and it was much higher in Na2CO3 treatment than in NaCl treatment at the same Na + concentration. The absorbance spectra and 4 th derivative spectra of photosynthetic pigments were declined under 300 mM NaCl and 25 mM Na2CO3 stresses, and remarkably changed under 50 mM and 100 mM Na2CO3 stresses. When the cells stressed by the two salts, the maximum quantum yield (Fv/Fm), electron transport rate (ETR) and photochemical quenching (qP) were reduced markedly, but the nonphotochemical dissipation (NPQ) was increased markedly. At the same Na + concentration, Na2CO3 stress had stronger toxic effects on photosynthetic ability than NaCl stress.
