In intensive aquaculture systems, high mortalities are frequently observed during the early life stages of marine fish. The aim of this study was to investigate differences in the susceptibility of turbot Scophthalmus maximus, halibut Hippoglossus hippoglossus and cod Gadus morhua to various strains of Vibrio anguillarum (serotypes O1, O2α and O2β), V. salmonicida and V. splendidus. The bath challenge experiments were performed using a multidish system, with 1 egg well -1. Unchallenged eggs and larvae were used as controls. Larvae in challenged groups that suffered high mortality rates were examined by immunohistochemistry. The overall results with respect to mortality showed that the O2α serotype was pathogenic to all 3 species, while the O1 serotype was pathogenic to halibut and cod. The immunohistochemical examinations revealed differences in histopathology. The O1 serotype produced more severe and highly developed infections than the O2α serotype. In larvae exposed to the O1 serotype, necrosis and bacterial cells were seen in the dermis, gastrointestinal tract, brain and eye area, while in larvae exposed to the O2α serotype, bacteria were usually limited to the gastrointestinal tract. These results suggest either that there are undetermined species differences in host immunity or that these pathogens are host-specific even in the early life stages of fish. The O2β strain did not cause an increased mortality to halibut and turbot.

Extensive genetic diversity has been discovered in the microbial world, yet mechanisms that shape and maintain this diversity remain poorly understood. This thesis investigates to what extent populations of the gamma-proteobacterial family, Vibrionaceae, are ecologically specialized by investigating the distribution across a wide range of environmental categories, such as marine invertebrates or particles in the water column. Additionally, it seeks to determine whether in situ population distributions directly result from a competitive advantage over other Vibrio populations. This was investigated by in vitro competition assays on mixtures of native, sterilized particles. Generalist populations were found to dominate the associations with marine invertebrates, consistent with a model of high migration dominated population assembly. A majority of populations occurred broadly within and among the different types of invertebrates sampled, with one population being a near perfect generalist with regard to seasons, host taxa and body regions. High variability across host individuals, consistent with a scenario of stochastic clonal expansion, was especially pronounced in crab and zooplankton samples. Specialization, demonstrated by specific and reproducible association with different particle types in the water column, is more common than specialization within invertebrate hosts. Co-existing Vibrio species show strong preferences for different types of particulate matter in the water column suggesting that competition for limited resources influences their evolution. While populations show different growth profiles on particle derived substrates, relative growth advantages of specialist populations in competition with other Vibrio populations on native particles may not be sufficient to explain observed environmental distributions. Instead, populations may gain an advantage on these particles by colonizing the living plant or zooplankton prior to death and degradation into particulate matter. In summary, although vibrios are known commensals of marine invertebrates, evidence suggests that population structure within animals is fairly weak compared to suspended particles in the water column. This highlights the importance of comparing multiple environmental categories and migration among them to investigate population structure and adaptation.

ABSTRACT. The use of live feed is common in aquaculture processes due to its nutritional advantages, and rotifers comprise one of the most used feeds in finfish aquaculture. However, bacteria present in water used for rotifer culture are a significant source of infection and consequently diseases could cause mortality in fishes. Advanced oxidation process (AOPs) are methods that may be useful to eliminate bacterial infections present in aquaculture wastewater and may allow the reuse of this water. Therefore, the prospect of using different advanced oxidation processes to inactivate Vibrio spp. and total bacteria cultures (TCB) present in this kind of water was evaluated. In particular, UVc/O3/H2O2, UVc/H2O2 and O3/H2O2 and the adsorption process were applied to seawater coming from a fish farming tank. The results obtained were highly promising because it was possible to inactivate 100% of bacteria present in the seawater when UVc/ozone/hydrogen peroxide and UVc/hydrogen peroxide were applied, needing around 20 and 80 min for Vibrio spp. and total culturable bacteria inactivation, respectively. These results are justified due to the strong action between H2O2 in the presence of UVc, which may produce a large amount of hydroxyl radicals. Nevertheless, when ozone/hydrogen peroxide was applied, total inactivation of bacteria did not occur. Also, an interesting relationship between bacteria inactivation and oxide reduction potential of seawater was observed during the process. Rotifers put in water after the treatment did not suffer behavior changes (swimming) and no mortalities were observed. El uso de organismos vivos como alimento es común en la acuicultura, debido a sus ventajas nutricionales. Los rotíferos constituyen uno de los alimentos principales en el cultivo de peces. Sin embargo, las bacterias presentes en aguas utilizadas en el cultivo de rotíferos son una fuente importante de infección y pueden causar enfermedades y mortalidad en peces. Los procesos de oxidación avanzados son métodos útiles para eliminar bacterias presentes en aguas residuales de la acuicultura, permitiendo su reutilización. Se evaluó la inactivación de Vibrio spp. y bacterias cultivables totales presentes en aguas residuales de la acuicultura a través procesos de oxidación avanzados. Particularmente, se estudió la aplicación de UVc/O3/H2O2, UVc/H2O2 y O3/H2O2 junto a procesos de adsorción aplicados a agua proveniente de un estanque de cultivo de peces.
