Abstract: Very small embryonic-like stem cells (VSELs) are a population of developmentally early stem cells residing in adult tissues. These rare cells, which are slightly smaller than red blood cells, i) become mobilized during stress situations into peripheral blood, ii) are enriched in the Sca1+Lin−CD45− cell fraction in mice and the CD133+ Lin−CD45− cell fraction in humans, iii) express markers of pluripotent stem cells (e.g., Oct4, Nanog, and SSEA), and iv) display a distinct morphology characterized by a high nucl… Show more

“…Danova-Alt et al [66] recently concluded that UCB VSELs neither have embryonic nor adult stem cell-like phenotype, are not equivalent to mouse VSELs, have aneuploid karyotype and should not be regarded as a stem cell population. However, they have studied Lin-/CD45-/CD34+ cells and not Lin-/CD45-/CD133+ cells, which are the VSELs as described by Ratacjzak and group [38]. Further it remains to be confirmed whether the aneuploidies they report are a technical artefact, since no cell lineage is expected to be aneuploid.…”

“…Ratajczak and group have made tremendous contribution to the field of VSELs biology. At present various laboratories across the world are providing evidence to support the pluripotent property and potential of VSELs isolated from cord blood and bone marrow [38]. Possible reason being the method to isolate VSELs by flow cytometry described by Ratajczak and group could easily be replicated in various labs across the world.…”

“…Thus, after two rounds of transplantation, the VSEL cells could have been as old as 8 months. Previous studies have demonstrated that VSEL cell loss occurs with increasing age [39]. Moreover, the serial transplant studies were performed in immunocompetent mice.…”

“…However, VSELs do not fulfill the in vivo criteria, as they do not complete blastocyst development and do not grow teratomas [25,81]. The reason for quiescence of these cells is the modification of expression of certain parentally imprinted genes.…”
