The GH growth axis plays an important role in the growth and development of animals and runs through the whole life of animals. Many studies have shown that molecular mutations in key genes of the GH axis will affect the growth and development of animals. The purpose of this study was to explore the distribution characteristics of InDels of GHR, GHRH, and GHRHR in seven Chinese sheep populations, and to further explore the relationship between InDels and sheep growth traits. GHR showed high variation in Chinese sheep, and GHR-53 showed the highest minimum allele frequency (MAF). There was only one InDel mutation site in both GHRH and GHRHR. The genotype frequencies of Hu sheep (HS), Tong sheep (TS), and Lanzhou fat-tail sheep (LFTS) were quite different from other breeds. The association between GHR, GHRH, and GHRHR InDels and body size traits in seven varieties were analyzed. The results showed that there was no significant relationship between GHRH and body size traits in the seven sheep populations. There was a positive association between GHR-21 and hip height of LFSH (p < 0.05). GHR-43 reduced body height and chest depth of Small tail han sheep (STHS) and hip width of TS. GHR-44 significantly affected the body weight of HS, the body height of STHS and the head depth of TS. GHR-53 significantly reduced cannon girth of HS, chest of STHS and forehead width of TS. GHRHR-2 significantly reduced the body weight of LFHS. To sum up, this study revealed the effects of GHR, GHRH, and GHRHR InDels on sheep phenotypic traits, which indicated their potential application prospects in the genetic improvement of mutton sheep.

Expression profile analysis to predict potential biomarkers for glaucoma: BMP1, DMD and GEM

Purpose Glaucoma is the second commonest cause of blindness. We assessed the gene expression profile of astrocytes in the optic nerve head to identify possible prognostic biomarkers for glaucoma. Method A total of 20 patient and nine normal control subject samples were derived from the GSE9944 (six normal samples and 13 patient samples) and GSE2378 (three normal samples and seven patient samples) datasets, screened by microarray-tested optic nerve head tissues, were obtained from the Gene Expression Omnibus (GEO) database. We used a weighted gene coexpression network analysis (WGCNA) to identify coexpressed gene modules. We also performed a functional enrichment analysis and least absolute shrinkage and selection operator (LASSO) regression analysis. Genes expression was represented by boxplots, functional geneset enrichment analyses (GSEA) were used to profile the expression patterns of all the key genes. Then the key genes were validated by the external dataset. Results A total 8,606 genes and 19 human optic nerve head samples taken from glaucoma patients in the GSE9944 were compared with normal control samples to construct the co-expression gene modules. After selecting the most common clinical traits of glaucoma, their association with gene expression was established, which sorted two modules showing greatest correlations. One with the correlation coefficient is 0.56 (P = 0.01) and the other with the correlation coefficient is −0.56 (P = 0.01). Hub genes of these modules were identified using scatterplots of gene significance versus module membership. A functional enrichment analysis showed that the former module was mainly enriched in genes involved in cellular inflammation and injury, whereas the latter was mainly enriched in genes involved in tissue homeostasis and physiological processes. This suggests that genes in the green–yellow module may play critical roles in the onset and development of glaucoma. A LASSO regression analysis identified three hub genes: Recombinant Bone Morphogenetic Protein 1 gene (BMP1), Duchenne muscular dystrophy gene (DMD) and mitogens induced GTP-binding protein gene (GEM). The expression levels of the three genes in the glaucoma group were significantly lower than those in the normal group. GSEA further illuminated that BMP1, DMD and GEM participated in the occurrence and development of some important metabolic progresses. Using the GSE2378 dataset, we confirmed the high validity of the model, with an area under the receiver operator characteristic curve of 85%. Conclusion We identified several key genes, including BMP1, DMD and GEM, that may be involved in the pathogenesis of glaucoma. Our results may help to determine the prognosis of glaucoma and/or to design gene- or molecule-targeted drugs.

We conducted a scoping review to 1) map and categorize published studies reporting interventions during the in vitro production of Bovine embryos and 2) qualitatively summarize the effects of treatments based on hormones, growth factors, sera, and reproductive fluids during oocyte maturation and embryo development. We searched electronic databases using keywords ('Bovine', 'embryo', 'blastocyst', 'oocyte', 'in vitro', 'quality') derived from the PIOS approach of the PRISMA statement. We identified 231 studies published during 2000−2021, with 133 being published between 2012 and 2021. The 231 studies were classified into four treatment categories: culture conditions (28), medium composition (45), bioactive supplements (79), and other additives (79), with 19 subcategories within these categories. A total of 77 studies included hormones, growth factors, sera, and reproductive fluids, among which 53 studies reported a positive effect on embryo production. Hormone-based treatments using melatonin, gonadotropins, and steroids were the most effective, followed by interventions assessing growth factors such as EGF, FGF, IGF-1, and BMP. These treatments improved oocyte competence, cytoplasmic/nuclear maturation, oocyte quality, and blastocyst development. Although free-serum media and synthetic/alternative products can partially or totally replace serum, it is sometimes necessary even at low concentrations. At high concentrations, hormones or growth factors may have detrimental effects on oocyte nuclear maturation, impair embryo development, and decrease embryo survival. Further meta-analyses are needed to estimate the intervention-specific effects. Future research should focus on interventions that have a positive effect and can be used extensively for Bovine production.
