Variation of gonococcal lipooligosaccharide structure is due to alterations in poly-G tracts in lgt genes encoding glycosyl transferases.

Abstract: SUlTlnlaryThe lipooligosaccharide (LOS) expressed by gonococci spontaneously varies its structure at high frequency, but the underlying genetic mechanism has not been described. We have previously reported that the genes encoding the glycosyl transferases responsible for the biosynthesis of the variable o~ chain of the LOS of Neisseria gonorrhoeae are located in a locus containing five genes, lgtA, lgtB, IgtC, IgtD, and lgtE. “…The lgtABCDE operon, originally described by Gotschlich, 21 encodes enzymes responsible for extension of the LOS a-chain, and three of the genes (lgtA, lgtC and lgtD) contain homopolymeric G nucleotide tracts that are subject to phase-variable expression. Single nucleotide deletions or insertions in these genes due to slipped-strand mispairing events can place these genes in or out of frame, 8,21 resulting in variable production of full-length and functional enzymes.
