Variant (6;15) translocations in murine plasmacytomas involve a chromosome 15 locus at least 72 kb from the c-myc oncogene.

Abstract: The variant (6;15) translocations in murine plasmacytomas join the myc oncogene‐bearing band of chromosome 15 and the immunoglobulin kappa band of chromosome 6. We recently cloned a region from chromosome 15 linked to C kappa and have now used probes from that region to define the major locus of plasmacytoma variant translocations, which we denote pvt‐1. In five of nine plasmacytomas we analysed, the 6;15 translocation resulted from reciprocal recombination between the C kappa locus and a 4.5‐kb region of pvt‐… Show more

“…With probe 8q305, we screened a genomic library, prepared from placental DNA in phage XEMBL3, and isolated a set of overlapping clones spanning a total of 30 kbp. Figure 3A shows a restriction map of the cloned chromosome 8 Cell line BL21 (27) harbors a complex translocation, t(2;8;9), in which, as judged by the banding pattern, a segment of chromosome 9 is inserted between the long arm of chromosome 8 and the translocated short arm of chromosome 2 on the 8q+ marker chromosome. To cloned size-fractionated HindIII fragments of BL21 DNA in phage XL47 and isolated clone 21.1 ( Fig.…”

“…Examples include observations that rearrangement of the region at 8q24 encoding MYC and PVT1 is frequently involved in human leukemias and lymphomas (4,5), the regions is frequently amplified in solid tumors (2), and a site of recurrent tumorigenic viral integration in mice (25). MYC is well established as an oncogene in this region.…”

“…The Pvt1 locus is also frequently targeted by proviral integrations in murine leukemia virus-induced T-cell lymphomas in mice (23) and in rats (24). It is also the site for breakpoints in ∼20% of human Burkitt's lymphomas (25) and in murine plasmacytomas (26). In conclusion, CAT thymocytes are a useful model to gain insights into the contribution of β-catenin to genomic instability because they sustain frequent illegitimate RAG recombination events leading to recurrent oncogenic translocations.…”
