Valproic Acid and Its Amidic Derivatives as New Antivirals against Alphaherpesviruses

Abstract: Herpes simplex viruses (HSVs) are neurotropic viruses with broad host range whose infections cause considerable health problems in both animals and humans. In fact, 67% of the global population under the age of 50 are infected with HSV-1 and 13% have clinically recurrent HSV-2 infections. The most prescribed antiherpetics are nucleoside analogues such as acyclovir, but the emergence of mutants resistant to these drugs and the lack of available vaccines against human HSVs has led to an imminent need for new ant… Show more

“… 2 Studies raise the possibility of the therapeutic anti-viral potential of VPA. 3 , 4 The virus replication and survival depend on the produced mitochondrial energy; thus, the anti-viral therapy tactics may involve medicine preparations that change energetic mitochondrial functions and activate anti-inflammatory mechanisms. 5 As a simple fatty acid, VPA is a substrate for the fatty acid β -oxidation pathway.…”

“…Antiviral natural products have gained immense consideration in herpesvirus research after the Nobel Prize in Physiology or Medicine (in 2015) was awarded for the discovery of the natural anti-malarial drug artemisinin. Thus, natural anti-infective agents might pave the road for defeating the problem of drug resistance as well as provide effective and safe treatment of infections caused by herpesviruses [ 13 , 14 , 15 , 16 ]. Curcumin, a natural phenolic compound found in the Curcuma longa L. ( Zingiberaceae ) (turmeric), could help eliminate certain viruses by various mechanisms of action ( Figure 1 ).…”

“…VPA has additional activities beyond inhibition of HDACs [17]. Clinically, VPA is a widely and frequently used anti-epileptic drug since 1963, owing to its activities in inhibition of GABA transaminase and blocking voltage-gated sodium channels and T-type calcium channels [18,19], and it has potential for the treatment of other neurological diseases, cancer and virus infection with unclear mechanisms [20][21][22]. VPA has been used to promote the differentiation of human stem cells into different types of somatic cells.…”
