Use of a multiplex polymerase chain reaction assay in the sex typing of DNA extracted from archaeological bone

Abstract: We developed an original method that allows the simultaneous molecular amplification and analysis of human chromosome X‐ and Y‐specific sequences from cortical compact bone using the polymerase chain reaction (PCR). The strategy is based on sequential amplification using external and internal primers. The targeted loci are SRY for chromosome Y and DXZ4 for chromosome X. Internal and external primer pairs for both loci were selected in order to allow the simultaneous amplification of short sequences of distinct… Show more

“…However, PCR products were not obtained in any tested sample, regardless of modifications of PCR conditions originally designed by Palmirotta et al [8]. In male samples, aDNA extraction was repeated four times and samples were subjected to DXZ4/SRY nested PCR amplification to exclude accidental occurrence.…”

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD). Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD. The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC). Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…Markers with an average heterozygosity ≥0.30 in different ethnic groups were also selected. The gender-typing loci DXZ4 and SRY were included in the study based on our previous results (12, 13) (Tab. I).…”

“…Likewise, many decontamination methods have been systematically tested for ancient DNA studies of skeletal remains. Some of these methods, such as scraping 31 and disk sampling 32 , have been used for permafrost decontamination as well. However, other methods used for aDNA studies of bone, including UV irradiation 33 , and treatment with household bleach 34 have not been tested on permafrost intended for microbial work.…”
