Use of 1H NMR STD, WaterLOGSY, and Langmuir monolayer techniques for characterization of drug–zein protein complexes

Abstract: Zein is a protein based natural biopolymer containing a large amount of nonpolar amino acids, which has shown the ability to form aggregates and entrap solutes, such as drugs and amino acids to form stable protein-drug complexes. In this work, π-A isotherm, NMR, and Dynamic light scattering were used to detect the formation of protein aggregates and the affinity between zein and two different drugs: tetracycline and indomethacin. An effective interaction of zein and the two drugs was evidenced by means of liqu… Show more

“…With an increase of PVA‐SbQ amount and a decrease of zein amount, the water contact angle was found to decrease. It was because zein has a more hydrophobic character due to more hydrophobic non‐polar amino acids than polar hydrophilic ones . A more hydrophilic nanofiber surface will be helpful for cells attachment and could provide a moist environment to accelerate the wound healing.…”

“…The water peak and TSP reference signal were also excluded. Spectra further aligned with the icoshift ( 73 ) algorithm and bucketed with the optimized bucketing algorithm ( 74 ), using a 0.005-ppm minimum bin size, leaving 1,652 data points per spectrum. These spectra were subjected to probabilistic quotient normalization (PQN) and Pareto scaled ( 75 ).…”

“…For these reasons, NMR spectroscopy is highly sought after in drug development [ 37 , 38 , 39 , 40 , 41 ], for both molecule identification [ 11 , 13 , 14 , 18 , 42 , 43 , 44 , 45 , 46 ] and structural elucidation [ 15 , 16 , 17 , 45 , 47 , 48 , 49 , 50 , 51 ]. NMR has been successfully applied in stereochemistry [ 52 , 53 , 54 , 55 , 56 ] and isomer determination [ 57 , 58 , 59 , 60 , 61 ], in drug-protein interactions studies [ 62 , 63 , 64 ], and in the evaluation of drug toxicity [ 65 , 66 , 67 , 68 ].…”
