Urease activity in soybean meal products

Abstract: SummaryA method which uses direct titration of ammonia as a measure of urease activity was modified for use in the assay of raw and of slightly denatured soybean meal. The modifications which were adopted were the use of glutathione, a presoaking of the sample for 30 min. at 40°C., and making of the assay at 40°C. Data on the urease activity of several varieties of soybeans, and for immature and frost‐damaged soybeans were determined.

“…The urease activity was determined by the titration method. 46 Briefly, 1.8 mg of urease was added to the buffer solution (3.6 mL, pH either 4 or 7) containing urea (30 mg/mL) in a test tube at 25 °C, with or without ADH (14 mg/mL) and/or PAM-co-PDAAM polymer (135 mg/mL). After 30 min, the enzymatic reaction was terminated by adding 30 mL of HCl (0.12 M).…”

“…One study [113] proposed an alternative method, with the potential to differentiate between meals with low levels of urease activity, based on the incubation of the meal with urea in a buffered solution and the colorimetric determination of the residual urea with pdimethylaminobenzaldehyde. A method for direct titration of ammonia as a measure of urease activity was proposed [114] and adapted [115], in which the incubation of urea and the meal is performed, maintaining the pH of the solution by slowly adding HCl. The system is then titrated with NaOH.…”

“…These values were on lower end of range of values reported for SBM samples from different geographical regions (55 samples, 70%-77%, Ravindran et al 2014;515 samples, 68%-96%, García-Rebollar et al 2016). The urease index indicates if applied heating is enough to destroy trypsin inhibitors (Smith et al 1956). The observed urease index was within the range of 0.00-0.25 reported for SBM from US, Brazil, India, and Argentina (Ravindran et al 2014;García-Rebollar et al 2016).…”
