Updates on Old and Weary Haematopoiesis

Abstract: Blood formation, or haematopoiesis, originates from haematopoietic stem cells (HSCs), whose functions and maintenance are regulated in both cell- and cell non-autonomous ways. The surroundings of HSCs in the bone marrow create a specific niche or microenvironment where HSCs nest that allows them to retain their unique characteristics and respond rapidly to external stimuli. Ageing is accompanied by reduced regenerative capacity of the organism affecting all systems, due to the progressive decline of stem cell … Show more

“…Comparing within each age group, the highest proportion of leukemia (35.5%) was seen among patients older than 50 years of age while there was a similar proportion between age groups 0-14 and 15-49 (32.3% each) (p < 0.0001). is high preponderance of older adults for leukemia may be mainly due to advancement of age where many environmental exposures to carcinogens, irradiations, and malignant mutations due to clonal expansion occur more frequently [27,28].…”

“…Interestingly, most studies on the role of BMA in the HSC niche have focused on their potential support of HSC survival/proliferation, with little attention to the possible facilitation of HSC differentiation. However, myeloid skewing is a common event in obesity, diabetes, myeloid malignancies, and aging [162]; all conditions with increased BMA. Considering that extramedullary hematopoiesis in adipose tissue is preferentially axed towards the myeloid lineage [163,164], the involvement of BMA in such a process in the BM under health and disease should be subject of future work.…”

“…The immune system's aforementioned diversity stems from it being comprised of various cells, all different in form and function. Despite its diversity, all cells of the immune system descend from the HSC niches in the bone marrow, which are tended to all cells of the osseous line, mesenchymal cells and endosteal cells (86). Depending on developmental factors secreted from these local cells, the stem and progenitor cells commit to different lines of evolution (Table 1) (85).…”
