Abstract: Although the development of abnormal myocardial mechanics represents a key step during the transition from hypertension to overt heart failure (HF), the underlying ultrastructural and cellular basis of abnormal myocardial mechanics remains unclear. We therefore investigated how changes in transverse (T)-tubule organization and the resulting altered intracellular Ca(2+) cycling in large cell populations underlie the development of abnormal myocardial mechanics in a model of chronic hypertension. Hearts from spo… Show more

“…For example, the association between early myocardial dysfunction and abnormal interstitial depositions of collagen and amyloid material has been elucidated in several animal models of HFpEF. 47,48 Similarly, in a rat model of chronic hypertension, it was recently noticed that progressive disruption of T-tubule and defects in calcium cycling started at an early stage and were associated with progressive subclinical reductions in systolic and diastolic myocardial mechanics. These changes preceded the occurrence of clinical symptoms and development of cardiac fibrosis, which was associated with the fall in LVEF.…”

“…The amplitude of this peak is called TT power and usually considered as the best indicator of the transversal organization level in the transverse-axial tubular system (2,10,11). Figure 1 represents the flow diagram of the TTorg algorithm.…”

“…These disorders impair contraction and may be responsible for some arrhythmias (11). Irregularity of TT has been described in atrial CM of several mammals (1) and occurs in a number of pathologies, including human heart failure (8,10), atrial fibrillation (5), and chronic ischemia (3). Therefore, the measure of the TT organization level represents an important issue for a better understanding of the physiopathology of these cardiac diseases.…”
