Ueber die Einwirkung von Aldehyden auf Orthodiamine der Pyrimidinreihe

Abstract

Einwirkung von Aldehyden auf O r t h o d i a m i n e der Pyrimidinreihe. [Aus dem pharmaceutischen Institut der UniversitLt Berlin.] (Eingegangen a m 11. Januar 1906.) Der Eine von uns hat vor einigen Jahren das Xanthin und riele diesem verwandte Verbindungen durch Eiuwirlrung o r g a n i s c h e r Sii u r e n auf gewisse o-D i a m i n 0-p y r i m i d i n e dargestellt I). Neuerdings haben wir gefunden, dass man zu Xanthinbasen auch gelangen kann durch Einwirkung VOII A l d e h y d c n auf dieselben Diamine des Pyrimidins in analoger Weise, wie man in der Reihe des Benzols dieden Purinkorpern in gewisser Beziehung rergleichbarensogenannten Anhydrobasen ebenfalls aus den Orthodiaminen entweder durch Einwirkung von Sauren oder von Aldebyden gewinnt. Wir haben unsere Versuche liauptaachlieh mit dem 3-Methyl-4.5diamino-2.6-dioxypyrimidin, dem 1.3-Dimethyl-4.5-diamino-2.6-dioxypyriinidin und dem 2.4.5-Triamirio-6-oxypyrimidin angestellt2). Giebt man zu der wassrigen Losung z. B. der zweiten Base einen Aldehyd wie B e n z a l d e h p d , so erfolgt sogleich Abscheidung eines Condensationsproductes, entstanden aus je einem Molekiil der Componenten uiiter Austritt eines Molekiils Waeser. Durch die leichte Spaltbarkeit bei der Einwirkung von Sauren wieder in Aldehyd und Diamin iat der Kiirprr als eine Renzylideuverbindung der Rase charakterisirt. Man kann nun diese Verbindung durch trocknes Erhitzen niit einem z w e i t e n Mo1.-Gew. Benzaldehyd in eine durch Saure nicht mehr spaltbare Verbindung iiberfiihren, die als ein im Glyoxalinring z w e if a c h snbstituirtes Purin, als 1.3-D i m e t h y l-7b e n z y l-8-p h e n y lx a n t h i n , aufgefasst werden muss: C H B. A Novel Synthesis of 3,9-Dialkyl and 8-Aryl-3,9-dimethylxanthines
