Tyrosinase-related protein (TRP)-2 is not only expressed on glioma cells, but is naturally processed and presented by their surface MHC molecules and is recognized by TRP-2-specific cytotoxic T cells. After active immunotherapy, we detected TRP-2-specific cytotoxic T lymphocyte (CTL) activity in patients' peripheral blood mononuclear cells (PBMC). Tumor cells from postvaccination resections showed significantly lower TRP-2 expression and higher sensitivity to carboplatin and temozolomide than those autologous cell lines from prevaccination resections in two patients who demonstrated CTL response to TRP-2. One of two patients underwent treatment with temozolomide after recurrence and responded dramatically. TRP-2-transfected cell line (TRP-2-U373) resulted in significant drug resistance to carboplatin and temozolomide compared to wild-type U-373 (W-U373). There was no significant difference, however, in the mRNA expression of other common drug resistance related proteins, such as BCRP-1, MGMT, MDR-1, MRP-1 and MRP-3, after TRP-2 transfection. TRP-2-U373 tumor cells were immunoselected by a TRP-2-specific CTL line. The immunoselected cells (IS-TRP-2-U373) demonstrated significantly increased sensitivity to carboplatin and temozolomide compared to TRP-2-U373. For the first time, we provide evidence that immunological targeting of tumor-associated antigen TRP-2 significantly increases sensitivity to chemotherapy.

The tyrosinase family comprises three members, tyrosinase (Tyr), tyrosinase-related protein 1 (Tyrp1), and dopachrome tautomerase (Dct). Null mutations and deletions at the Tyr and Tyrp1 loci are known and phenotypically affect coat color due to the absence of enzyme or intracellular mislocalization. At the Dct locus, three mutations are known that lead to pigmentation phenotype. However, these mutations are not null mutations, and we therefore set out to generate a null allele at the Dct gene locus by removing exon 1 of the mouse Dct gene. Mice deficient in Dct [Dct tm1(Cre)Bee ] lack Dct mRNA and dopachrome tautomerase protein. They are viable and do not show any abnormalities in Dct-expressing sites such as skin, retinal pigment epithelium, or brain. However, the mice show a diluted coat color phenotype, which is due to reduced melanin content in hair. Primary melanocytes from Dct knockout mice are viable in culture and show a normal distribution of tyrosinase and tyrosinase-related protein 1. In comparison to the knockout, the slaty mutation (Dct slt /Dct slt ) has less melanin and affects growth of primary melanocytes severely. In summary, we have generated a knockout of the Dct gene in mice with effects restricted to pigment production and coat color.

Purpose: Chemotherapy-resistance of melanoma has led to poor prognosis and decreased survival in the patients. Therefore, the addition of adjuvant therapies to the conventional chemotherapy regimens has been taken into consideration to improve the clinical treatments efficiency. In this study, the effect of microwave (MW) Hyperthermia has been evaluated on the toxicity of cisplatin on the MM200 cell line in the presence and without gold nanoparticles (GNPs). Methods: Cells incubation was performed with and without cisplatin in the presence and absence of GNPs. To induce hyperthermia, the cells were immediately placed under MW irradiation for 25 and 30 minutes (41-43°C) following the addition of the drug and GNPs, then they were incubated for 24 hours. Finally, cell survival was determined by MTT assay.Results: GNPs (up to 6.6 µg/mL) showed no toxicity. GNPs at the concentration of 13.2 and 26.4 µg/mL caused 13% and 20.7% drop in cell survival rate, respectively. IC50 of cisplatin decreased from 4 to 2 µg/mL in the presence of GNPs. Hyperthermia (43°C) plus chemotherapy (2 µg/mL) resulted in no significant enhancement in cisplatin cytotoxicity relative to chemotherapy alone whereas by adding GNPs, an increase in cell mortality up to 15-fold in comparison to cisplatin alone was observed. Conclusions: There is a synergistic effect between cisplatin and GNPs, this could be due to the facilitated entrance of cisplatin in the presence of GNPs. MW exposure improves the efficacy of cisplatin therapy in the presence of GNPs on MM200 cells.
