Two-photon microscopy imaging of oxidative stress in human living erythrocytes

Abstract: Red blood cells (RBCs) are known to be the most suitable cells to study oxidative stress, which is implicated in the etiopathology of many human diseases. The goal of the current study was to develop a new effective approach for assessing oxidative stress in human living RBCs using two-photon microscopy. To mimic oxidative stress in human living RBCs, an model was generated followed by two-photon microscopy imaging. The results revealed that oxidative stress is clearly visible on the two-photon microscopy imag… Show more

“…The oxidative stress inside living RBCs was determined via the two-photon laser scanning microscopy imaging described earlier [ 81 , 82 , 88 ]. RBCs were isolated from the fresh blood samples using isotonic phosphate-buffered saline solution [ 81 , 82 , 88 , 89 , 90 , 91 ] and used for two-photon microscopy imaging immediately to avoid any alterations in the erythrocyte morphology.…”

“…Common organic fluorophores, such as fluorescein derivatives, have been modified with ROS-sensitive moieties that quench fluorescence; however, on ROS-mediated cleavage they possess fluorescence suitable for TPEF (26). Furthermore, the team of Lewis recently developed an azulene fluorophore for TPEF bioimaging of RNS and ROS (27).…”

“…Oxidative status is generally assessed in erythrocytes, tissues, and plasma; however, because erythrocytes are exposed to biochemical damage and can be easily collected, they are frequently analyzed . Oxidative stress in erythrocytes has also been the focus of research in cats with DM.…”

The activity of superoxide dismutase in erythrocytes is lower in diabetic cats than in controls and does not increase during treatment.…”
