Two N-Linked Glycans Differentially Control Maturation, Trafficking and Proteolysis, but not Activity of the IL-11 Receptor

Abstract: Background/Aims: The cytokine interleukin-11 (IL-11) has important pro- and anti-inflammatory functions. It activates its target cells through binding to the IL-11 receptor (IL-11R), and the IL-11/IL-11R complex recruits a homodimer of glycoprotein 130 (gp130). N-linked glycosylation, a post-translational modification where complex oligosaccharides are attached to the side chain of asparagine residues, is often important for stability, folding and biological function of cytokine receptors. Methods: We generate… Show more

“…Similar to IL-6R, the ectodomain of IL-11R is subjected to N-linked glycosylation. However, in contrast to IL-6R, glycosylation at Asn-194 seems to be essential for the transport to the plasma membrane and substitution of Asn-194 with alanine resulted in predominant localization to the ER ( Agthe et al, 2018b ).…”

“…Nglycosylation is initiated in the ER, and the sugar chains are then further processed in the Golgi. Therefore, the fully matured IL-11R at the plasma membrane has a higher molecular weight compared to intermediates still located in intracellular organelles [36,38]. Additionally, the ER has an important role in terms of quality control, as it retains not correctly folded proteins and prevents their onward transportation to the Golgi.…”

“…After translation, the IL‐11R is transported through the endoplasmic reticulum (ER) and the Golgi apparatus before being inserted into the plasma membrane. The amino‐acid sequence of the IL‐11R contains two sequons for N ‐linked glycosylation at the two asparagine residues N127 and N194 [38]. N ‐glycosylation is initiated in the ER, and the sugar chains are then further processed in the Golgi.…”
