Two laccase isoforms of the basidiomycete Cerrena unicolor VKMF‐3196. Induction, isolation and properties

Abstract: The laccase induction in submerged culture of basidiomycete Cerrena unicolor VKM F-3196 was investigated. Cu(2+) at concentration 0.1 mM was an optimum inducer of C. unicolor laccase. Two isoforms of laccase, namely LacC1 and LacC2, were isolated and characterized. The isoforms were shown to have different physical-chemical and catalytic properties. On the basis of the MALDI TOF MS analysis of tryptic cleavage products of both the proteins and N-terminal amino-acid sequences analysis two isoforms of laccase (L… Show more

“…Among them, copper and aromatic compounds are considered as the most potent inducers of enzyme synthesis (Revankar and Lele 2006; Elisashvili and Kachlishvili 2009; Lisova et al 2010; Janusz et al 2013). In this study, supplementing the culture medium with 1 mM CuSO 4 or 1 mM xylidine significantly accelerated laccase secretion and increased enzyme yield (Figure 2).…”

“…There are few reports on the production of LME by the Cerrena unicolor strains (Janusz et al 2007; Lisova et al 2010; Songulashvili et al 2012; Hibi et al 2012; Rola et al 2013). However, the knowledge related to the physiological regulation of C. unicolor LME synthesis is limited to develop economical ways of industrial scale enzyme production.…”

“…The molecular weight of the laccase from S. commune was reported to be 62 KDa by SDS-PAGE [De Vries et al, 1986], suggesting the carbohydrate content to be around 13.7%. The variations in the experimental findings could be due to the presence of many laccase isoforms with different characteristics [Dong et al, 2005;Giardina et al, 2007;Lisova et al, 2010]. Slight variations in the expressed peptide sequences are attributed to differences in patterns of glycosylation in P. pastoris contributing to the differences observed.…”

“…The laccase-inducing ability of guaiacol is observed in many basidomycetes, but there is not sufficient evidence of the molecular mechanism of laccase regulation by an inducer. [16,37,38] In the static culture, 8 times more laccase was obtained using 0.906 mM guaiacol added at 96 hr of fermentation compared to the enzyme obtained without using any inducer (without inducer, laccase activity 26 U=L). For the same concentration and time of addition of guaiacol, 40 times higher laccase production was observed in the shake culture than that obtained without any inducer (without inducer laccase activity $4 U=L) (cf.…”
