Two-Dimensional Gel Electrophoresis Analyses of pH-Dependent Protein Expression in Facultatively AlkaliphilicBacillus pseudofirmusOF4 Lead to Characterization of an S-Layer Protein with a Role in Alkaliphily

Abstract: The large majority of proteins of alkaliphilic Bacillus pseudofirmus OF4 grown at pH 7.5 and 10.5, as studied by two-dimensional gel electrophoresis analyses, did not exhibit significant pH-dependent variation. A new surface layer protein (SlpA) was identified in these studies. Although the prominence of some apparent breakdown products of SlpA in gels from pH 10.5-grown cells led to discovery of the alkaliphile S-layer, the largest and major SlpA forms were present in large amounts in gels from pH 7.5-grown c… Show more

“…Therefore, pH 6?0 and 9?0 were used for comparative gene expression studies. Similar pH values have been used to study pH-stress conditions in E. coli K-12 (Maurer et al, 2005), Bacillus pseudofirmus (Gilmour et al, 2000) and Shigella flexneri (Small et al, 1994). An adaptation phenomenon was observed when cultures grown at pH 9?0 were reinoculated at pH 10?0.…”

“…In particular, alkaliphilic Bacillus spp. have been shown to produce secondary wall polymers that associate with the peptidoglycan layer and presumably are able to bind cations and enhance their availability for pH homeostasis (1,14). These bacteria also have high levels of squalene in their membranes, and it has been proposed that the high levels of this lipid might lower the permeability of protons across the lipid bilayer (18).…”

“…Although a B. pseudofirmus OF4 MgtE conferred significant Mg 2ϩ transport capacity at pH 7.0 in a heterologous system (32), pH optima of MgtE proteins may be higher in the native alkaliphile membrane. Several examples of such hard-wired adaptations to extreme alkaliphily have been found to disadvantage B. pseudofirmus OF4 at near-neutral pH (20).…”
