Two-color SERS microscopy for protein co-localization in prostate tissue with primary antibody–protein A/G–gold nanocluster conjugates

Abstract: SERS microscopy is a novel staining technique in immunohistochemistry, which is based on antibodies labeled with functionalized noble metal colloids called SERS labels or nanotags for optical detection. Conventional covalent bioconjugation of these SERS labels cannot prevent blocking of the antigen recognition sites of the antibody. We present a rational chemical design for SERS label-antibody conjugates which addresses this issue. Highly sensitive, silica-coated gold nanoparticle clusters as SERS labels are n… Show more

“…In practice, the conventional bioconjugation technique does not provide site-specificity and there is no control over targeting a particular amino acid residue. This uncontrolled binding in terms of missing site-specificity may result in a "scrambled" orientation of the antibodies on the surface of the SERS nanotags ( Figure Unauthenticated Download Date | 6/7/19 1:39 AM 2A) [13]. Thus the antigen recognition sites of the antibody (FAB) cannot recognize its target perfectly.…”

“…Such stabilizers are able to bind with NPs covalently, like peptides or IgGs due to their free amine residues [9,13]. Figure 2B shows the competitive binding of IgGs and stabilizers with NP.…”
