Abstract: Two cyanine-based fluorescent probes, ( E)-2-(4-(diethylamino)-2-hydroxystyryl)-3-ethyl-1,1-dimethyl-1 H-benzo[ e]indol-3-ium iodide (L) and ( E)-3-ethyl-1,1-dimethyl-2-(4-nitrostyryl)-1 H-benzo[ e]indol-3-ium iodide (L), have been designed and synthesized. Of these two probes, the twisted-intramolecular-charge-transfer (TICT)-based probe, L, can preferentially self-assemble to form nanoaggregates. L displayed a selective turn-on fluorescence response toward human and bovine serum albumin (HSA and BSA) in ∼100… Show more

“…BSA is as erum transport protein that contains severals ites that can bind aw ide variety of organic and inorganic species. “…To first characterize the optical properties of the fluorophore moiety in the most general setting (i.e., without a targeting ligand), we studied the intermediate probes 1 a , b and 2 a , b using bovine serum albumin (BSA). BSA is a serum transport protein that contains several sites that can bind a wide variety of organic and inorganic species [38–40] . In particular, it has been reported to bind fluorogenic triphenylamine derivatives of our compounds [41] .…”

“…Additionally, we studied the fluorescence behavior of 4 in different water-glycerol fractions and observed fluorescence enhancement by increasing the solvent viscosity (Figure 5b). Several twisted intramolecular charge transfer (TICT)-based fluorogenic probes for various targets, such as biothiol, reactive oxygen species, and HSA, demonstrated the TICT characteristics of the probe through theoretical calculations, showing the charge separation in the HOMO-LUMO transition, and by experimental confirmation of the solvent effect on fluorescence [47,48,49,50,51,52,53]. As in previous reports, these results, including the ICT character and viscosity-dependence of the fluorescence intensity of 4, showed the typical phenomena of a TICT-based fluorogenic probe.…”

“…Unlike the undesirable ACQ phenomenon, the aggregation of AIE dyes improves fluorescence, which expands the effective range of a probe. 23 Therefore, the typical AIE molecules B2-A2 and B3-A3 were applied for BSA detection and quantification.…”
