TRIM32 Promotes the Growth of Gastric Cancer Cells through Enhancing AKT Activity and Glucose Transportation

Abstract: Tripartite motif protein 32 (TRIM32), an E3 ubiquitin ligase, is a member of the TRIM protein family. However, the underlying function of TRIM32 in gastric cancer (GC) remains unclear. Here, we aimed to explore the function of TRIM32 in GC cells. TRIM32 was induced silencing and overexpression using RNA interference (RNAi) and lentiviral-mediate vector in GC cells, respectively. Moreover, the PI3K/AKT inhibitor LY294002 was used to examine the relationship between TRIM32 and AKT. Quantitative reverse-transcrip… Show more

“…The protective effects of AC-YVAD-CMK against short-wavelength blue light were investigated. The concentration of AC-YVAD-CMK was determined by a CCK-8 assay (SAB, USA) [26]. The HLE-B3 LECs were grouped into 3 groups as follows: A: the control group, which consisted of hLECs cultured in the dark; B: the caspase-1 inhibitor group, which consisted of HLE-B3 hLECs treated with AC-YVAD-cmk (20 μmol/L); and C: the short-wavelength blue light exposure group.…”

“…Experimental illumination systems (455-460 nm) were installed on top of the cell culture incubator as previously described. We adopted an illuminance of 2500 lx [26] to study the potential influence of blue LED light on hLECs.…”

“…Proteins and hormones in distinct cancer can utilize PI3K to promote glycolysis. For example, Ubiquitin-specific protease 6 Nterminal-like protein sustains chronic AKT phosphorylation and GLUT1 stability fueling aerobic glycolysis in breast cancer ( 177); TRIM32 promotes the growth of gastric cancer cells by enhancing AKT activity and GLUT1 expression (178). Studies have discussed that PI3K-induced glycolysis may be responsible for the formation of chemoresistant phenotypes of cancer cells (20), and the inhibition of glycolysis by interrupting the PI3K signaling pathway can automatically improve chemoresistance.…”

“…In contrast, overexpression of TRIM32 improved the phosphorylation of Akt and inhibited apoptosis. TRIM32 is expressed in all cells of the human body and, therefore, may be a general regulator of growth in normal and cancerous tissues that require a rapid increase in cell size [ 141 ]. A better understanding of TRIM32 function may lead to therapeutic interventions to either increase cell size in patients with mutations in TRIM32 or to limit the growth of cancers that overexpress TRIM32.…”
