Trends in SPR Cytometry: Advances in Label-Free Detection of Cell Parameters

Abstract: SPR cytometry entails the measurement of parameters from intact cells using the surface plasmon resonance (SPR) phenomenon. Specific real-time and label-free binding of living cells to sensor surfaces has been made possible through the availability of SPR imaging (SPRi) instruments and researchers have started to explore its potential in the last decade. Here we will discuss the mechanisms of detection and additionally describe the problems and issues of mammalian cells in SPR biosensing, both from our own exp… Show more

“…Indeed, in SPR imaging, signal results from amplitude modulation at a fixed angle, unlike GC-SPR where a phase angle scan is performed. Because they are based on intensity interrogation, SPR imaging sensors suffer from worse performance with respect to angle scanning systems ( [20] and for a complete review see [21]). In conclusion, our methodological approach results in good resolution, compact, inexpensive and very simple cell detection setup, and opens for further research strategies based on the integration of the sensitive grating into a microfluidic chamber that could improve cell biology studies.…”

“…With each well containing a single cell, and most of the cell's secreted species diffusing towards the SPR surface rather than the bulk, the ability of SPRi instruments to take measurements at multiple detection spots enables a comparison of the secretions of different cells [156]. For more details, the reader is referred to a recent review on the advances in the field of SPR cytometry [157].…”

“…This relatively large sensing depth allows SPR to monitor the changes occurred not only on surfaces, but also in the bulk. SPR has been applied in various studies for molecules with molecular weight between 1000 Da~500 kDa [47]. Liu et al [48] attached human ovarian carcinoma SKOV-3 cells on the ceiling of a flow cell chamber, and detected their vascular endothelial growth factor (VEGF) secretion by monitoring the SPR signal of the antibodiesimmobilized gold chip at the bottom of the flow cell.…”
