Trazodone Hydrochloride: A Wide Spectrum Antidepressant with a Unique Pharmacological Profile

Abstract: Trazodone is a new antidepressant agent that was recently introduced in the United States. It has a unique pharmacological profile that is not typical of either tricyclic or monoamine oxidase inhibitor antidepressants. As such it represents a new class of antidepressant drugs. The efficacy of trazodone has been clearly established in comparative studies with imipramine and amitriptyline. Major depression is the principal indication for its use, but good results have been shown in a wide variety of depressive s… Show more

“…Trazodone is an antidepressant agent with a chemical structure unrelated to existing psychomotor drugs (Al-Yassiri et al, 1981;Brogden et al, 1981;Ayd & Settle, 1982;Georgotas et al, 1982;Rawls, 1982) which is sedative and impairs critical flicker fusion threshold in young and elderly volunteers (Bayer et al, 1983).…”

“…From these and other data [24], trazo done seems to produce fewer anticholinergic side effects than the tricyclic antidepressant agents, although this is usually only statisti cally significant for the complaint of dry mouth. Drowsiness does occur with trazo done, but not to the extent of producing an appreciable dropout rate, as compared to that seen with mianserin.…”

“…Trazodone is an antidepressant agent with a chemical structure unrelated to existing psychoactive drugs [AlYassiri et al, 1981;Brogden et al, 1981;Ayd and Settle, 1982;Georgotas et al, 1982;Rawls, 1982] which is sed ative and impairs critical flicker fusion threshold in young and elderly volunteers [Bayer et al, 1983], The present study was designed to examine the phar macodynamic effect and pharmacokinetics of trazodone in comparison to a standard sedative antidepressant, amitriptyline [Hindmarch, 1980], alone or in combina tion with a 'social dose' of ethanol to assess possible interactions between the agents.…”
