Transplanted human cord blood-derived unrestricted somatic stem cells improve left-ventricular function and prevent left-ventricular dilation and scar formation after acute myocardial infarction

Abstract: Transplantation of USSCs significantly improved LV function and prevented scar formation as well as LV dilation. Since differentiation, apoptosis and macrophage mobilisation at infarct site were excluded as underlying mechanisms, paracrine effects are most likely to account for the observed effects of USSC treatment.

“…This was associated with a significant improvement of left ventricular ejection fraction and it prevented left ventricular dilation. 67 We were not able to confirm USSC graft survival and USSC differentiation toward cardiomyocytes. However, with 10 8 USSC used for engraftment in the study by Kim et al, the cell number exceeded ours significantly (1.2 Â 10 7 ).…”

“…Patient's lifelong exposure to risk factors contributing to ischemic heart disease (ie, age, diabetes mellitus, and smoking) may impair the potential of autologous stem cells. [11][12][13] Restricted availability is present because selection and culturing of sufficient potent cells is cumbersome and time consuming. This limitation is especially important in the acute setting of myocardial ischemia.…”

“…USSC have been shown to enable reconstitution of femoral defects in mice 3,5,52 and to improve heart function after myocardial infarctions. 3,7,53,54 In this study, mesodermal differentiation capacity of the clones was confirmed by the ability of the cells to mineralize after culturing in medium that induces stem cells down the osteogenic differentiation pathway. Distinct clusters of Alizarin Red S-stained cells were observed in both clones confirming successful mineralization.…”
