Abstract: Tumor metastasis is one of the big challenges in cancer treatment and is often associated with high patient mortality. Until now, there is an agreement that tumor invasion and metastasis are related to degradation of extracellular matrix (ECM) by enzymes. Inspired by the formation of natural ECM and the in situ self-assembly strategy developed in our group, herein, we in situ constructed an artificial extracellular matrix (AECM) based on transformable Laminin (LN)-mimic peptide 1 (BP-KLVFFK-GGDGR-YIGSR) for in… Show more

“…Based on this observation, several groups are developing NPs to mimic and reinforce ECM in the TME to inhibit metastasis. For example, to prevent tumor invasion and metastasis, a transformable laminin-mimic peptide (BP-KLVFFKGGDGRYIGSR) was generated [151]. The BP motif of the peptide could self-assemble onto NPs due to the strong hydrophobic interactions.…”

“…Because of the similarity with natural laminin, the peptides on the NPs could readily transform into nanofibers around the solid tumor upon binding to integrins and laminin receptors to form an artificial ECM [151]. The results indicated that the artificial ECM remained at the tumor site for a couple of days which significantly inhibited the lung metastasis in breast and melanoma tumor models [151]. The clinical utility of such therapeutic strategy remains to be tested in relevant clinical settings.…”

“…The overlap region of the two spectra (Figure b) was more than 30 %, which indicated the possibility of the FRET mechanism occurring . BP was modified with a carboxyl group, according to our reported method, which endowed BP with a negative charge to interact with positive RB. Negative BP nanoaggregates were synthesized before being coated with RB‐PAH.…”

“…To evaluate the invasive ability of SiHa and HeLa cells, wound healing and invasion assays were performed . The migration healing ability of SiHa cells was defined as the 100% wound healing rate at 24 h after scratching (Figure C); the wound healing rate of SiHa cells was much higher than that of HeLa cells (50.58%, Figure D).…”
