Leptospirosis in sheep is often underestimated, and leads to great economic losses for the sheep farming industry. The aim of this study was to evaluate the humoral immune response in pregnant ewes, after the injection of a commercial polyvalent vaccine for leptospirosis, and to observe the transmission of anti-Leptospira antibodies through the colostrum to the offspring. For this, 24 pregnant ewes were vaccinated for leptospirosis. Blood samples were collected prior to vaccination and then 7, 14, 21, 28, 35, 42 and 49 days after vaccination. In order to evaluate passive immunity transfer, blood samples of 32 lambs were collected during the first 48 hours after birth, and another collection was performed 10 to 21 days after birth. The lambs were placed into 2 groups: Group A (n=16): singleton lambs; and group B (n=16): twins. The sera samples were submitted to the Microscopic Agglutination Test (MAT), in which 21 Leptospira serovars were tested. The results were analyzed in a descriptive form. The number of sheep reactive to MAT gradually increased until 21 days after vaccination, and decreased right after. Of all the serovars contained in the vaccine, the largest proportion of animals were seroconverted to Hardjoprajtino serovar, Serjoe serogroup. Anti-Leptospira antibodies transferred through colostrum to lambs were detected by MAT in the serum collected 24-48 hours after birth. It was observed that 65.6% (21 out of 32) of the lambs were reactive. In the subsequent collections that occurred from 10 to 21 days after birth, a decrease in the number of animals reactive to the MAT was detected. There was no significant statistical difference for the passive transfer of antibodies between single or twin lambs.

O objetivo deste estudo foi a comparação das metodo-logias mais comuns empregadas em diferentes fazendas para a avaliação da qualidade do colostro e identificação de falha na transferência de imunidade passiva (FTIP) em bezerras Holandesas. Sessenta e oito bezerras Holandesas recém-nascidas foram distribuídas em dois grupos: G1 - provenientes de propriedade com manejo de colostro adequado; G2 - oriundas de propriedade com falhas no manejo do colostro. Foram colhidas amostras de colostro fornecido para cada bezerra de cada grupo (250 ml) para a avaliação de IgG, sólidos totais, PT e albumina. Amostras sanguíneas foram obtidas às 48 horas de vida para a determinação de PT e de globulina e para a estimativa indireta de IgG por meio dos testes de turvação pelo sulfato de zinco, precipitação pelo sulfito de sódio e coagulação do glutaraldeído. Os teores de IgG observados no colostro pelo colostrômetro (G1: 98,63 ±1 5,58 mg/ml; G2: 85,66 ± 29,81 mg/ml; p = 0,09), índice Brix (G1: 27,61 ± 2,96%; G2: 22,40 ± 4,58%; p = 0,00), PT (G1: 10,42 ± 2,29g/dL; G2: 9,04 ±2 ,29g/dL; p = 0,06) e albumina (G1: 1,25 ± 0,37g/dL; G2: 0,91 ± 0,38g/dL; p = 0,00) foram superiores no G1 quando comparados ao G2. A determinação de PT no soro das bezerras foi realizada por um analisador bioquímico automático, revelando taxas de FTIP (< 5,2 g/dL) somente no G2 (46,67%). Os resultados dos testes qualitativos (turvação pelo sulfato de zinco, precipitação pelo sulfito de sódio e coagulação do glutaraldeído) foram diferentes dos resultados de PT (p = 0,00) e não se mostraram eficientes para a detecção da FTIP. Esta pesquisa demonstrou que além do uso do colostrômetro para a avaliação da qualidade do colostro, a utilização do refratrômetro Brix e níveis de PT do colostro devem ser consideradas. A identificação de FTIP no rebanho pode ser realizada seguramente por meio dos níveis séricos de PT e globulina.

In agrarian reform settlements there are obstacles in the rearing of calves; however, the main aspects that cause this obstacle are not known. Thus, the aim of this study was to diagnose the main obstacles in the management of these dairy heifers to target extension actions. For this, semi-structured guided questionnaires were applied to 40 families from 20 settlements. The questionnaire contained questions about the characteristics of the family, property and general management, feeding and sanitation of calves. The data were tabulated in Excel, transformed into variables and studied by means of multivariate analysis of main components after the farms were grouped into clusters according to similarity. The variables that most affected the heifer rearing system in dairy farms in the agrarian reform settlements of Santana do Livramento, RS, Brazil, were related to diet and occurrence of diarrhoea in calves. The 40 farms studied were grouped into six clusters, which differed especially in relation to who was responsible for the management of the heifers inside the property, regarding the infrastructure for the calves, time in milk production and concern regarding the care in the breeding of the calves. Based on these differences between the clusters being correctly identified, the technicians who work in these farms can establish strategies to work with settled families, emphasising the points where the obstacles are known to trigger the survival of heifers that will replace the dairy herds.
