Transcription factor YY1 functions as a PcG protein in vivo

Abstract: Polycomb group (PcG) proteins function as high molecular weight complexes that maintain transcriptional repression patterns during embryogenesis. The vertebrate DNA binding protein and transcriptional repressor, YY1, shows sequence homology with the Drosophila PcG protein, pleiohomeotic (PHO). YY1 might therefore be a vertebrate PcG protein. We used Drosophila embryo and larval/imaginal disc transcriptional repression systems to determine whether YY1 repressed transcription in a manner consistent with PcG func… Show more

“…The coincidental increase in H3K27me3 (Fig 3) suggests that the recruitment of Polycomb group complex (PcG) might contribute to establish marks of repressive chromatin, in cooperation with Brm-based SWI/SNF, as proposed by Ho & Crabtree [51]. Interestingly, the sequence of Ccnd1 promoter that is bound by Brm and is enriched in H3K27me3 contains both putative YY1 and E2F4/6 binding sites, which can mediate Brm recruitment, via PcG [52] or pRb [49,53], respectively.…”

“…The search site predicted a series of transcription factors that might be involved in the transcriptional regulation of let-7a (Table 1). The transcription factor Yin Yang 1 (YY1), ranked first in the prediction, is a member of the polycomb group and has been shown to be involved in CXCR4 signaling (25)(26)(27)(28)(29).…”

“…One mechanism of YY1-mediated transcriptional repression is through recruitment of PcG proteins (44,45). In situ hybrid- knockdown experiments (sets 1 and 2) using whole cell extracts of untransfected (Ϫ) or transfected P19 cells with either pSUPER-YY1 (YY1 siRNA) or pSUPER-CTRL (ctrl siRNA) vectors.…”
