Transcription Factor Achaete Scute-Like 2 Controls Intestinal Stem Cell Fate

Abstract: The small intestinal epithelium is the most rapidly self-renewing tissue of mammals. Proliferative cells are confined to crypts, while differentiated cell types predominantly occupy the villi. We recently demonstrated the existence of a long-lived pool of cycling stem cells defined by Lgr5 expression and intermingled with post-mitotic Paneth cells at crypt bottoms. We have now determined a gene signature for these Lgr5 stem cells. One of the genes within this stem cell signature is the Wnt target Achaete scute… Show more

“…As shown in Fig. 3g, blebbistatin stimulated the expressions of the pluripotency marker genes Lgr5 and Ascl2 in Lgr5 þ intestinal stem cells 2,28 . To further support the notion that blebbistatin promotes the survival and pluripotency of Lgr5 þ stem cells via Akt activation, we examined whether the PI3K-specific inhibitor LY294002, which can block Akt activation 29 , could attenuate the effects of blebbistatin.…”

“…Such uncoupling between Lgr5 and Olfm4 expression was puzzling as Olfm4 was described as a robust marker of Lgr5-positive ISCs. 25 To clarify this point, we first looked at Olfm4 expression by in situ hybridization. Most crypt bases were devoid of Olfm4 expression, and consisted of packed lysozyme-positive paneth cells (Figure 2e Figure S1B, arrow).…”

“…The genes included Paneth cell markers ephrin receptor B3 ( EphB3 ) and lysozyme 1 ( Lyz1 ) (van Es et al ., 2005) or genes with broader expression in various crypt cells such as division cycle associated 7 ( Cdca7 ), ephrin receptor B2 ( EphB2 ), naked cuticle homolog 1 ( Nkd1 ), and SRY (sex determining region Y)‐box 9 ( Sox9 ) (Batlle et al ., 2002; Schuijers et al ., 2015; Stancikova et al ., 2015). Remarkably, the most robust downregulation was recorded for three genes that encode Wnt signaling‐activated markers of ISCs achaete‐scute complex homolog 2 ( Ascl2 ), Lgr5 , and Troy (Fafilek et al ., 2013; van der Flier et al ., 2009). Interestingly, olfactomedin 4 ( Olfm4 ), which represents another ISC marker but whose expression is not regulated by Wnt signaling (van der Flier et al ., 2009), was decreased only moderately.…”

Finally, expression of keratin 20 ( Krt20 ), which marks terminally differentiated cells, was at the first time point substantially increased, but at the later time point returned to its original level (Fig.…”
